Thanks for posting this! It’s good to hear that other supervisors and senior/lead techs have gone through the same convoluted paths. It is encouraging to hear that I’m not the only older adult that is back in school while working a full time job!
Flotation bath temperature depends a little on personal preference. It is usually 5°C to 10°C below the melting point of the paraffin. If the paraffin used melting point is 58°C, then the water bath should be OK in the range of 48°C-53°C. Sections are not "exploding".
Hi, I hv tried sectioning rodent brains on a thermoscientific HM525 NX. I have tried sectioning in a range of -15 to -22 . Adjusted the clearance angle to 10,11,15,9. Nothing seems to be working. The sections are badly folded or stick to the anti-roll plate. At -17 a few sections come here and there. But m in dire need of seriel sections. However, the machine works well for rodent
