I think u have to use the plant Nuclei extraction you have to use tris HCl 0•4 M pH 7.5, always with ice bath and filter pores 20 micrometers this protocole is used for Allium cepa roots
Our Nuclei Extraction Buffer (130-128-024) has been validated for a wide range of fresh and frozen mouse and human tissues. We have not tested the Nuclei Extraction Buffer on fungal culture, I am afraid. However, we think that the buffer may be applicable to a wide range of species.
During a manual magnetic cell separation, the negative fraction will drip by gravity and it is necessary to perform the 3 washing step and to collect them as well as part of the negative fraction. What is this machine doing? Vibrating the cells? Shaking? What's the approximate rotation area and rpm? What is this machine doing that just shaking the cells at high RPM is not doing?

I understand that when Car-T therapy is used for chronic Lymphocytic Leukemia,it wipes out all B-cells,what about NK-cell therapy?Would it leave the heathy B-cells?

NK cell-based therapeutic strategies in chronic lymphocytic leukemia (CLL) and in other malignancies aim to potentiate and/or restore NK cell activity or to provide patients with functional NK cells able to kill tumor cells. According to Sportoletti et al (NK Cells in Chronic Lymphocytic Leukemia and Their Therapeutic Implications - PMC (nih.gov)) ‘’in vitro studies have shown that allogeneic NK cells, after expansion/activation with an optimized protocol, are able to kill CLL cells independently of KIR-HLA mismatches. However, although allogeneic NK cells do not mediate the GvL effect in a CLL-HSCT transplantation setting, they, unlike T cells, lack the potential to cause graft-versus-host disease (GvHD) thus representing appealing and safe candidates for adoptive immunotherapy for CLL.” Your Miltenyi Team
