I didn't have an interpreter on the day I did this lab in my class, and I was so lost! I was nervous to find out that this process was going to be on exam! Yikes! However, I finally understand thank you! The closed caption helped me significantly, again thank you SO MUCH! :)

Hope you guys pursue biology as a major/minor in college, because, in high school, you study the experiments and what happens, as well as what we now understand. But in college, you get to actually perform these experiments and reinforce what you studied in high school and relearned in college!

5 out of 30 students were successful in growing transformed bacteria? Psshhh. No big deal. My class had 100% of the students successfully grow bacteria that glowed. Again, no big deal. We be swaggy like that.

1:20 . For some reason ,for plate 4 , almost 90% of the lawn glow when shined with UV light .. which contract because it is supposed to glow in clusters . So what went wrong ? Anyone know ??

GFP is just a protein, but plasmids are DNA. DNA has to be transcribed into mRNA and then translated into a functional protein. The gene for GFP is useless if there is not a starting point for transcription called a "promoter"--that is the araC that you see in the video. AraC is an inducible promoter, meaning that it is usually off but the presence of some chemical (arabinose) can turn it on. Thus, arabinose will turn on the promoter, which initiates transcription of the GFP gene.

Beneficial is relative to the context of the selective pressures of the environment. A common transformation protocol involves the solution mentioned in the video alongside a heat shock, which will shock the bacteria and "trick" them into uptaking dna. What is not mentioned is that bacteria can get rid of useless plasmids during replication, which is why it is important that these plasmids for lab work have a selectable marker (such as one that encodes for resistance to an antibiotic).

CaCl2 is a “divalent cation.” Can you explain why a “monovalent cation” like NaCl would not be as effective?

The reason why divalent cations (basically always Ca++) work is because they can simultaneously be attracted the negatively charged bacterial membranes and the negatively charge DNa. This allows Ca++ to basically bring the DNA closer to the cell. Monovalent cations would only be able to be attracted to one or the other.
