PCR stands for polymerase chain reaction (kary Mullins in 1985).It is a technique of production of multiple copies of recombinant DNA invitro using DNA polymerase enzyme. Working Mechanism Of PCR= This reaction involves three basics steps. 1.Denaturation 2.Annealing. 3.Primer extension. Denaturation=In this Target DNA is heated to a very high temperature (94 degree Celsius). Heating results in the separation of two strands of Target DNA. Each of two strands of Target DNA then acts as template for the synthesis of new DNA strand. This step is called denaturation. ANNEALING=In this two oligonucleotide primers join to each of single standard template DNA. The sequence of primers is complementary to 3' end of template DNA.Annealing is carried out at lower temperature (40-60 degree Celsius). PRIMER EXTENSION - During this step enzyme DNA polymerase synthesize the DNA segment between the two primers. Usually Taq DNA polymerase enzyme isolated from a thermophillic bacteria thurmus acquatics is used in most of the cases. The two primers extends towards each other to copy the DNA segment. This step requires deoxyribonucleoside triphosphate and Mg2,+ and occurs at 72 degree Celsius temperature. These three steps complete the first cycle of PCR and two copies of DNA are formed.For Second cycle these two DNA acts as Target DNA.
