... no? there hasn't been any point in the game that says "choose Frisk's/Chara's gender" and while debatably maybe you can interpret Frisk's to be whatever you want, the fact that Chara is only addressed by neutral/nonbinary pronouns still remains even if Chara may not be nonbinary, sometimes pronouns have more so to do with masculinity/femininity than sex; for example, i myself am a male and do not view myself as nonbinary or something similar yet i still want to be addressed with they/them
