How can we decide in which ISO class our clean room should include? We don't know particle count of room.

For that you need to do qualification of that room and based on the outcome of the result you can classify that room

ISO 14644-1:2015 is not a guideline, it is a standard. Also F.F.209E is not English standard it is an obsolete 10 year old American standard.

For example if your area is grade B and the in operation limit for grade B area is considered for the at rest limit of grade C. IN OPERATION LIMIT OF ANY AREA = One grade down limit.
