Cloning a chimera - mushroom anomaly! (two mushrooms one bag?)

In this video we are going to clone the wierd anomaly of two oyster mushrooms growing in the same bag - is this a genetic mutation or two mushrooms fighting for the same turf? 🍄❤️

49 years of breeding and I learn more from this kid than I could ever teach him. Mycology? bout 10 blocks in my life time and some mushroom adventures with Denver's botanical garden peeps. lol, all these years reading and pronouncing the word "chimera" wrong, lot to be said bout college, btw horticulture will be a kwel fun class where as math english BS so Im not here to correct the kid. plants are my subject and white strips on leaves can be a genetic type in the strain called variegation like he refered to or the white strip could be a mutation as well. I THINK when he says that the this shroom anomaly could be the result of stress he referring to a phenotype and not a genotype and cloning should/could help determine this., but not sure if that what he was saying as far as pheno vs geno or a mutation of a new genome. Awesome vid and cant wait to see the results and his procedures..

Thanks for watching! I am definitely still learning as well but enjoy exciting moments like this - it might be a contaminated oyster from another oyster but it all appeared to be from the same cluster 👍 the plates are performing well so we shall see! 🍄❤️

I had 2 blocks of what are supposed to be Blue Oysyer mushrooms. But both blocks looked like the light brown color oyster mushrooms. I thought maybe fresh air, or humidity was why in my case. Will be finding out soon

What do you think of adding activated carbon to your plates for cloning ? In my case it really seems to help to reduce contaminations.

You mean, you mix the activated carbon into the agar? Do you only do this during cloning or do you use it for other purposes as well?

@@johnandjacquelinewarren9979 Yes mix it with the agar while liquid. I used it while cloning, but I guess it would be useful anytime you have a risk of contamination. I don't remember where I first read about it. But I don't know if my results were the result of better lab practices the one time I used it or if it really helps. That's why a side by side comparaison would be interesting.

@@BIBIwood Thanks. I am just getting started, but am trying to research as much as I can and love finding ideas like this. Mixing in a bit of carbon seems like a pretty easy step if it is beneficial. But like you said a side-by-side test would be nice to see if it is more than just better lab practices.

​@@BIBIwood charcoal is known to be detoxifying and to have antimicrobial properties! It's just unknown for sure (through rigorous lab testing) whether those antimicrobial properties extend to the main microbes we deal with in mushroom cultivation -- though I imagine it would be effective even against those, and based on anecdotal evidence from cultivators online, the consensus seems to be "yes, charcoal works". How well it works, and what concentration of charcoal is required, and whether charcoal can be added to other parts of the process aside from agar, are all unknown to me at this time. I'd like to experiment to see if adding charcoal to grain spawn and bulk substrate would also help cut down on contamination risk. 😊

Those plates there.. are they for use in the future when needing to make xfers? And are they parafilmed I can’t tell

Those are freshly poured plates that he keeps in his hood until further use like cloning, QC of LC, spore germination etc. I don't think they are parafilmed, that's why they are kept in the hood.

I assumed "yes", since he leaves the plates there unwrapped... but at the beginning of this video, he specifically said "I'm going to use the flow hood today for the light", which seems to imply to me that it was off before the video... which means the plates were sitting under the hood area, but without the sterile air flowing over them. I hope he corrects me if I'm wrong. I'd very much like to know if blank plates can be left at room temp UNwrapped and in a non-sterile environment, or if they need to be kept in a sterile environment if UNwrapped. Knowing this will help me reduce plastic waste (and especially once I move to the much more expensive Parafilm, I'd very much like to cut down on my use lol).

they ended up fizzling out before they fruited so Im not sure- went back to the OG slant and this never occurred again 🤷

It can be mutation, but i guess, this is just spore-contaminated substrate. The first non-natural oyster species has found on a random mushroom. If the clone drops more chimeras, we have the answer. I guess, you may have a consultation with a biologist, who can explain the situation, and identify the other variation of oyster mushroom. Good luck!
