Nope, not real. Well actually every note and every phonetic sound is sung by a choir and recorded. This way you can let them sing what you want and let them sing texts while doing so. It is an absolutely phenomenal technology that is only improving!

was this created with separate samples, or is it an original composition with sheet music?

It's composed in a DAW, it doesn't work with literal notes but with MIDI notes. The sounds are samples, each note of each instrument is recorded separately.
