This is so helpful. I start my new job as a histology technician in five days. I’m excited and trying to find as many videos on the process so I’m not completely lost on my first day

How is this the exact cryostat we have at my lab? This is great, thank you for the video. An impatient lady who didn't speak English trained me so I've been calling things "the-thing-that-holds-the-sample" and so forth lol, I'm glad to hear the actual terminology.

many missing points: 1) collecting sections with brush (I think it's very important to handle this part properly, because so many wrinkles are on sections because of bad methods of collecting. 2)lab coat 3)touching the door, the fridge and everywhere with gloves 4) that gray garbage bin is not used for tissue samples. it had to be bio-hazard container with the special sign on it and also with a proper lid. 5) it's better that OCT covers the top of sample too.

Do they teach the brush technique for obtaining sections? I've observed it often works better, but it does take some skill to get a good section.

Good, but a few things missing. Never mentioned about the angle of the stage, each tissue is cut at different temperature and thickness and why is he not wearing a lab coat?

I'm pretty new to cryosectioning but I guess this video is not meant for people with a proper lab training. First of all, you should consult the handbook before you even think of taking the sample off the freezer. Same goes for reading any protocol before starting an experiment, refresh your memory before doing anything (literally anything!), unless you know it by heart and do it regularly. If you think you know it but haven't done it for half a month - refresh you memory! Then, label your slides before you start cutting. While it does not seem nesessary if you only cut one sample, if you make a habit of cutting things and marking them afterwards, one day you may find yourself in a big of trouble. 2:45 well, 1) you'd better get rid of this section than putting it on a slide; and 2) "collect as many samples as possible on your slide" - the video contradicts the statement, there are 2 sections there and I see the space for a third whereas there could be at least 4 if you're new and really careful and 5-6 if you know what you're doing but do not go extremes. This garbage bin does not look like a special biohazard bin, doesn't have any sighs on it nor it has a proper lid. Another thing they don't teach you in a grad school is that you may disregard PPE in some cases. And it's not taught for a good reason. You have to understand good enough what you're dealing with before you do that. Cutting human or even animal samples is never the case. Apart from that and quite a few other remarks, it's a great tutorial from students for students.
