38:58 mam..vibrio cholerae produce yellow colonies not black colonies. The non Vibrio spp produced H2S and hence the colonies appeared black; on Thiosulfate Citrate Bile Salts Sucrose (TCBS) agar, colonies of Vibrio cholerae typically appear as yellow, smooth, and slightly translucent colonies with a distinct morphology.

The yellow coloration is due to the fermentation of sucrose by Vibrio cholerae, which produces acid and lowers the pH of the agar, resulting in the yellow color change of the pH indicator present in the medium.

In simple media all the basic nutrient such as peptone, beef extract, water, electrolyte basically present that gives basic nutrient. Simple media may be liquid or solid Synthetic media is such defined media in which we already know it's all basic composition of nutrient which you provide. Complex media is undefined in which it's chemical composition are not known . For eg nutrient broth, Macoonkey agar, tryptic soy agar are the some example of complex media
