Human Blastocyst Trophectoderm Biopsy preformed by RGI.
Cells collected from the trophectoderm. The force of the surface tension on the boundary of the biopsy drop and the mineral oil helps to separate the TE cells from the blastocyst. Approximately 5 cells collected from the outer layer of the embryo (trophectoderm) for preimplantation genetic testing (PGT).

I am a doctor worked in the reproductive medicine center of First-class Hospital at Grade 3 in China, I like this video so much. As RU-vid is blocked from accessing in China, If you don’t mind, I would like to share this video to other website in China for the purpose of popularizing knowledge of reproductive medicine and patient education. Of course, I will state the original author information at the end of the video. Thank you very much

We extend the tip to elbow length and we do this in-house with specialized equipment.

Impressed technique, how bigger hole laser for assisted hatching on day4? would you freeze embryo immediately after biopsy operation?(within 30min) thanks for your kindly response.
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 2 года назад
The hole can be 5-10 µm. Yes, freezing immediately after biopsy is acceptable.

We use custom pipettes with a longer tip to elbow distance for added control

Are you using this technique with every biopsy? Or do you apply the flicking technique as well? And regarding the technique show in the video, when you go to the edge of the drop, do you apply any suction from the biopsy pipette or do you just pull with the holding as I think you're doing in this particular biopsy? Thank you
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 2 года назад
At RGI we try to use the edge of drop for most embryo biopsies. This approach allows us to make the cytoplasmic bridge between the sample and embryo as thin as possible. That, in turn, allows us to minimize the number of laser pulses to 1-2 or avoid them completely. To stretch the cytoplasmic bridge to its maximal we do both: pulling with holding pipette and pulling by the biopsy pipette simultaneously. We also use flicking, but in rare cases, when for some reason a laser cannot completely detach the biopsy sample from trophectoderm.

And lastly, do you perform the assisted hatching on D3 or D5 or do you just wait for normal hatching? Sorry for the many questions.
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 2 года назад
In this video, assisted hatching was performed on day 4. Hatching can also be performed on the day of the biopsy on day 5/6.

Do you perform biopsy on blastocists on day-7 or you just discard after the 6th day of colture?
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 2 года назад
We do not discard potential embryos on day 6 and will preform day 7 biopsy on good quality embryos only.

0:52 Don't you afraid that you might drag all of the cell out of the zona when you are pulling ?
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 7 месяцев назад
No, we use the edge of the drop specifically to prevent the embryo from being pulled out of the zona.

@@reproductivegeneticinnovat8725 I tried your method but the drop did not cut off the cell, it just make my embryo become into strip shape.
@reproductivegeneticinnovat8725
@reproductivegeneticinnovat8725 5 месяцев назад
In this video, we used a single laser pulse (400 micro seconds @ 100% power) that resulted in the cells separation from the trophectoderm. Without laser assistance we would not be able to remove the cells. The edge of the drop is used only to stretch and thin out the bridge between the trophectoderm and biopsy cells with allows us to minimize the number of laser pulses.

Per PGDIS guidelines, mosaic transfer recommendations are deferred to the provider. In our PGT lab, where we work with clinics from around the world, mosaics account for about 8% of PGT-A diagnoses.

is this biopsy necessary? and this cell will be sent to mother back or another one? i just dont understand how this cell will live after you cut a good amount of it...
@reproductivegeneticinnovat8725
Only about five cells are removed from the outer layer of the embryo. This layer will eventually become the placenta and therefore should not affect the development of the embryo. This technique is necessary for PGT.
