Most stunning cooling I have seen on a GPU. I would love to embark on something similar. My worry would be leakage and poor installation that would result in hissing, gurgles or coilwhine.

Usually, the water in a water-cooled build needs to be changed about once a year, depending on the usage of the PC. The temperatures typically idle around 30°C, the processor reaching about 70°C, and the GPU at around 60°C under workload.
Its ok. Just maintain the water level above the inlet hole of the pump. If the water level drops below the inlet, it will cause noise and reduce the flow of water.
