In molecular biology, cloning a gene means inserting the sequence of the gene into a plasmid, which is a small piece of circular DNA that can be "transfected" into a bacterial strain, where the plasmid will replicate to many, many copies. The plasmid carries an antibiotic resistance gene, so when the bacteria are grown in the presence of the antibiotic, only those bacterial cells with the plasmid in them, can multiply.
This allows molecular biologists to easily propagate and grow vast amounts if the gene, and also to manipulate it in many ways, for example to introduce mutations to study its function, etc.

The answer to question no 11 is Option C. From the map we can see the the total size of the vector along with the insert is 3000+2000=5000bp(1kb=1000bp). When BAM H1 and Hind III cuts the construct we get one fragment of 500bp as it was told in the question that the insert has a Hind III site at 500bp downstream from its first codon and the Bam H1 site is at the beginning of the insert. So we get a band at 500bp and another band at 5000-500=4500bp=4.5kb which is in option C. And in case of Hind III and Pst1 digestion we first get a 1500bp(1.5kb) length band as Hind III site is 500 kb upstream of the first codon and Pst1 site is at the end of the insert. And the total length of the insert is 2000bp. So the band length should be 2000-500=1500bp=1.5kb. The other band will be of 5000-1500=3500bp=3.5kb in length. In the options C justifies all these. So C is the right option.
