Mamure Kalesi, zamanın ötesinden gelen bir miras, 3. yüzyılda Roma İmparatorluğu döneminde inşa edildiği tahmin ediliyor. Ancak daha sonra Bizans, Selçuklu ve çeşitli dönemlerde onarımlar ve eklemeler yapıldığı bilinmekte.

Mamure Kalesi, stratejik bir konumda bulunması sebebiyle tarih boyunca çeşitli medeniyetlerin ilgisini çeker. Savunma amacıyla görevini yapar ancak aynı zamanda bir garnizon ve ticaret merkezi olarak da kullanılır.
Özellikle Anadolu Selçuklu Sultanı I. Alaeddin Keykubat döneminde önemli bir onarımdan geçer ve o dönemdeki görünümünü büyük ölçüde kazanır. Kalenin adı da bu dönemdeki yapımına atıfla "Mamure" olarak değişir.
