I recently got the Espeon plush from build a bear and I'm not happy with the half inch "line" for the nose they factory stitched on, this is a very helpful tutorial to give my plush a proper sniffer! she has no long fur, so it may be a challenge, but I feel confident now. Yes, it would work with embroidery floss. It will take longer, but it would work. I don't think regular sewing thread would work very well. But... if you don't have any other choice then it is certainly worth trying. A skein of embroidery floss can be purchased for less than $1 at JoAnn's Fabrics.
If you have threads from a friendship bracelet weaving kit, I’ve found that those can work, too, since they are similar. You’d have to get a needle with an eye big enough to thread through, but most needle kits are $2 or so, sometimes cheaper at places like Dollar General or something.

This video is very helpful to me since unfortunately my dog managed to chew on my plushie's nose, I'll probably start in a few days but for now do you have any tips on how to sew a hole shut before making the nose because my dog kinda ripped it (also I'm planning to give this toy to a special someone! I'm sorry that your dog chewed your plushie! It depends on where the hole is located. If the hole is at the tip of the muzzle where the nose was, then I'd first try to add some more stuffing through that hole. Then I'd whip stitch the hole closed with a thread that matches the fabric backing as close as you can. then I would stitch the nose.
Normal thread has a different texture. If you are having trouble finding pearl cotton, you can find it online. If you prefer to buy locally then you could try embroidery floss and build that up.
