Thanks for watching. I'll recommend to go for steam-distillation instead of hydro-distillation as yield and quality it would be more efficient

Thank you Kiran for watching the video. You should go for hydro-distillation if you want ocimum leaf oils. Shade dry the leaves, crush with hands (do not grind in grinder as it may cause loss of volatile components due to friction). Soak the crushed leaves in water (in the same RBF) overnight for better yield.

Thanks for the video sir, it's tough to separate the water content from oil. If I add anhydrous sulphate then it's become impure and 2ndly in our we don't have fractional distillation unit in our lab. So, what's the good or alternative method sir, please sir give your valuable suggestions

Thanks for watching. Collect the distillate (with water), shake with CCl4 or CHCl3 in a separatory funnel. Collect the lower layer from the separator and evaporate to dryness at room temp in a graduated tube.

No need to use a fractional distillation unit. Use a separating funnel and shake distillate with diethyl ether (caution;: hingly inflammable) then evaporate ether layer

Sir, I was taught that during hydro distillation process there is an extraction medium, like a non polar solvent (hexane) is used to absorb the gaseous oil as it flows from the condenser. But you didn't made mention of it. Is hexane important in hydro distillation extraction?

Yes, you are taught correctly. If the extracted oil is very less or if the oil is to be used for any phytochemical analysis, then we make use of any water immiscible solvent (hexane, pentane or ether) to dissolve it. Later on we may separate the layer & evaporate the solvent. But, if we just want to quantify the oil (for QC of crude drug) separation is not necessary. I appreciate your querry. Thanks for watching.

There's no specific ratio. It depends upon the flask. Water is just a heating medium, else the drug will get charred.

Hello sir, I need your suggestion..we use steam distillation for extraction of Essential oil of Eucalyptus and pine ....we get essential oil but excessive amount of water in it.. further we use distillation unit for the separation of water...but we didn't get essential oil.

Many thanks for watching. Instead of distillating the oil-water mixture, just let it cool in refrigerator. The oil will float on water, then you may separate by separator (separating funnel) only if the quantity is sufficient. Alternatively, you may shake the oil-water mixture with diethyl ether n separate the ethereal layer and evaporate to dryness at room temp.

Hello Priya, thanks for watching. Betel leaf does not contain sufficient essential oil. However, steam distillation is always more efficient than hydrodistillation for any crude drug.

If you have the assembly for steam distillation, I'll suggest to go with it. As steam distillation is advanced technique and has more yield, reduced hydrolysis of essential components. An electric rice cooker with slight modification (for steam collection) can be least expensive device. Of course, you'll need a professional for modification.

Is Hydro Distillation better for making hydrosols? I guess yes cause in this way the essential oils stay into the hydrosol? I read that when hydrosols is a by-product of essential oils like in steam distillation the hydrosol is not so good in quality. I want to make the best quality of hydrosols... I know that for essential oils steam distillation is way better... Please Dr let me know !

Hydrosols are usually made up of floral oils. Most of the floral essences are thermo-labile (degrade at high temperature). Hence, hydro-distillation is not the preferred method. Enflurage method is ideal for floral essential oils. Finally, these isolated oils can be mixed in a co-solvent (few mls of ethanol) then dispersed in water. For citrus oils you may use eculle method/solvent extraction. If possible go for Supercritical fluid extraction it is ideal for all kinds of oil.

Ex. Suppose you have taken 25 g of clove powder & you've got 1 ml oil. The % yield = (100 × 1)÷ 25 = 4 % v/w Where v represents oil in ml obtained & w represents crude drug taken in g

Its RBF a short form for round bottom flask. A heating mantle has complimentary pocket to accommodate an RBF and heat efficiently.

Preserve in tightly stoppered container in a cool and dark place, preferably in an amber bottle. Some oils may darken upon exposure to light. You may refrigerate some oils as well (4-10 degree) Thanks for watching

Hello Mansi, Thanks for watching the video. Let me first clarify that water is a heating medium in this experiment. It will keep on circulating through the return tube. If the crude drug has sufficient amount of thermostable volatile oil, then, you may find it either on the top or bottom of the water column in the receiver. Initially check the powdered crude drug for presence of volatile oil by pressing it between 2 filter paper, it should show oily stain (that evaporates on heating). Alternatively, you may gently heat a pinch of crude drug with water in a test tube, it should evolve aromatic odor. Check n let me know if these tips are helpful.
