Hi, nice instruction! I have a question, is the reference cell always filled with de-ionized water?

Hi, the reference cell should be filled with the same solvent used in sample cell. In most cases, it is filled with milli-Q water.

Hi, usually we don't change the reference cell. Only in rare cases you may want to fill the reference cell: 1. You need to use a different solvent in the sample cell. (Never happened in this lab because we always use water as solvent) 2. You are troubleshooting the errors in your data that might be caused by contaminations in reference cell. (Never happened in this lab. This is very unlikely because as you can see at 2:35, the hole to the reference cell is extremely small, and in normal procedure you won't be able to put any contaminations into reference cell) If you want to learn more about ITC and reference cell, please check the user's manual. Hi, you should first import (read data) both data for baseline and titration results. Then, go to the deltaH window, double click the "1" icon on the top left of the graph, put both baseline and titration data on the same graph (the same way you plot multiple data sheet in the same plot in origin lab). Next, click "subtract baseline" button, and select the right baseline data and titration data, click "OK", you should get the subtracted data. If you did all the steps correctly, the end of the curve should be close to zero on the y axis. Thanks.
