thanks for the explanation. I've seen Neurofibromatosis disease mechanism today and learned about the neurofibromin protein (NF1 gene) but couldn't understand its function at first...now I see that neurofibromin is GAP that stops MAPK/ERK signaling pathway. thanks a lot

hello, there is something I dind't understand... does this pathway, just like AKT pathway and STAT pathway, happen in all immune cells, such as tissue-resident cytotoxic T cells (IE-CTL)?

Just woww But I would like to give a note : The Ras is active only when it is attached to the membrane. When Ras is in a free formule in the cytosol, then it is inactive. It is just a remarque. Thanks for all from France 😁✌🏻

You are correct. Recruitment of RAF by RAS actually facilitates the dephosporylation of ser259 which results in the disruption of the interaction between RAF and inhibitory 14-3-3 proteins, therefore activating RAF. RAS also facilitates RAF activation by driving RAF dimerizaiton.

I like this video, but Ras does not phosphorylate Raf. Instead, it binds it, and that is what activates it. Most cell biology textbooks like Molecular Biology of the Cell or Lodish Molecular Cell Biology explain it this way with cited experimental assays as evidence.

Excellent video Hussain. I think one only important thing missing from the picture @3:12 is what phosphorylates RAF. From another video (ru-vid.com/video/%D0%B2%D0%B8%D0%B4%D0%B5%D0%BE-ql62kMK3BrM.html) what I learnt is that a scaffolding protein binds RAF (which is bound to Ras) as well as Src kinase to form a complex. The scaffolding protein increases the local concentration of Src and RAF allowing them to interact. Src then phosphorylates RAF to fully activate it. I just subscribed your channel to learn more basic biology. Many thanks as you are doing a great job!

KInda of topic, but if you want to be precise with your drawings please draw the nuclear membrane with four layers of phospholipids, not like cell membrane that has only two layers.
