In this video, Biology Professor (Twitter: @DrWhitneyHolden) will cover the Oxidase test procedure and discuss the background information to understand how this test works. Specifically, this diagnostic test indicates the presence of cytochome c oxidase in a bacterial culture. This is an enzyme that participates in the electron transport chain during aerobic respiration. This test is used to differentiate oxidase-positive bacteria, like Pseudomonas and Neisseria spp., from oxidase-negative bacteria.

Do the aerobic bacteria testing -ve on the oxidase test lack the cytochrome c oxidase ?

Yes, they often have another kind of oxidase to allow electron transport for aerobic respiration

@@BiologyProfessor So the (TMPD) reagent is only oxidized by the cytochrome c oxidase NOT the other alternative oxidases .

That is more or less correct, I think. Check out this paper. The last few sentences of the abstract summarize the other types of cytochrome oxidases in oxidase-negative bacteria. @@BiologyProfessor That paper adds more questions than answers ! Mammalian cytochrome c oxidase (a , a3) is present in the majority of the bacillus strains testing -ve on the oxidase test . So the (TMPD) oxidation might be associated with another set of oxidative enzymes ; NOT necessarily those of the aerobic respiration .

@@BiologyProfessor According to what we got from that paper ; the (TMPD) acts as an electron donor to cytochrome c in order to let that cytochrome proceed towards the final step of the respiratory chain catalyzed by the cytochrome c oxidase enzyme . In a way the (TMPD) simulates the role of reduced coenzyme Q (CoQH2) in complex III of mammalian respiratory chain . Lord knows best !
