Yes, but ultimately you get better results if the ink it up against the PPP (photopolymer plate). This allows less light in, from the thickness of the transparency film. This is why it is done this way. It is probably negligible but again, I show the best way to get the absolute best results.

lanetxgp1: You will get better results if you are always putting emulsion or ink face down on the TNF or PPP. This is why it was printed in reverse and then flipped over so you could read the words face up. The least amount of light and space you have between the letters, the better the results because there is no UV light that leaks on the edges during processing.
