Neurotoxicity is a key side effect of the CAR-T therapy axicabtagene ciloleucel (axi-cel). Preclinical data has suggested that the IL-1-targeting anakinra can help prevent neurotoxicity. Paolo Strati, MD, The University of Texas MD Anderson Cancer Center, Houston, TX, discusses primary analysis results from a pilot study (NCT04432506) of prophylactic anakinra, aiming to mitigate CAR-T associated neurotoxicity in patients with relapsed or refractory large B-cell lymphoma (R/R LBCL). Dr Strati explains the study design of this trial and reports that the use of anakinra reduced neurotoxicity by approximately 50%.
