How is the amount of solvent determined? Thank you, I’m having problems with a batch I think I added too much water to the flask

@momsmushroomsjodyfoster5786 There are two aspects: 1) The solvent volume should be high enough so that the flask does not run dry when the solvent evaporates and fills the Soxhlet well. The solvent volume should be low enough to allow evaporation. Our suggestion: At the start, fill the Soxhlet well manually with solvent to just below the apex of the siphon and fill the flask to the level of its maximum diameter. 2) Depending on the compound amount to be extracted, you should not oversaturate the solvent in the flask, which would cause separation (e.g. precipitation). If you see oversaturation, use less botanical material in the Soxhlet well. Have you considered ultrasonic extraction as an alternative or add-on to Soxhlet extraction, yet? Thank you so much for your fast response! It’s difficult finding answers for my questions! But, again please. Should I be adding 250mills of liquid to the a 500mill flask? Because the liquid expands as its heated, correct? I tried adding more than 250 mils of water, and it was disastrous.

@momsmushroomsjodyfoster5786 Fill the flask to just above the widest diameter. Depending on the volume of your Soxhlet apparatus, you may have to add a little more, if you see the level in the flask falling too low. Make sure that your condenser condenses all water vapor, so that you don't lose water during the process. You see that our Allihn condenser has a very narrow opening on the top to avoid vapors exiting instead of condensing. Use a low coolant temperature on the condenser. The expansion of liquid due to heat is insignificant. Use boiling stones to get smoother boiling in the flask and use medium heat to start slow. Once you have everything under control, you can increase heat in small steps to speed up the process. Alternatively use cold water and sonication and avoid the Soxhlet all-together. In a Soxhlet extraction, the extracted compounds are exposed to heat in the flask for a long time, causing heat degradation. With our sonicators, you can extract at cold temperatures, so that the compounds are not exposed to heat.

Thank you for the great video! I was wondering what is the difference between Soxhletation and Distillation, and this video greatly helps with that! But I'm still wonderibg, why was it called "Cold Extraction" when in fact, the analytes was heated along with the solvent...?

@dimdimbramantyo7666 the Soxhlet extraction from the sample is „cold“, because the condensed solvent is cold. And yes, the heat exposure in the flask can be problematic for many heat sensitive compounds. That is why our ultrasonic probes enjoy great demand in botanical extraction. You can use our sonicators at very cold temperatures, e.g. 30 degrees below zero for solvents. This preserves heat sensitive compounds and produces great extraction yields and quality. Fantastic video but I was wondering. Are there no other alcohol soluble compounds in tobacco that you would collect along w/ the nicotine?( I stepped away from a moment. So maybe I missed that explanation. Or maybe it's something obvious i'm just not thinking of)

@etownKeystoned Yes, there are other alcohol-soluble compounds in tobacco that would be extracted alongside nicotine in a Soxhlet extraction. While nicotine is a key target compound, tobacco contains a variety of other constituents that are also soluble in ethanol. In addition to nicotine, tobacco contains other alkaloids like nornicotine, anabasine, and anatabine, all of which are soluble in alcohol. Also, polyphenolic compounds, responsible for the flavor and aroma, are generally alcohol-soluble. Ethanol can dissolve terpenes, which are aromatic compounds also present in tobacco. Phenols and other similar structures, which may contribute to tobacco's characteristic smell and taste, can be extracted as well. For this Soxhlet demonstration, ethanol acts as a broad-spectrum solvent, and its use in Soxhlet extraction will collect not just nicotine but a range of other alcohol-soluble components present in tobacco. Hello. I would like to ask. If I would want to extract mushrooms, is it better to use soxhlet extraction setup or sonicator? With soxhlet to make water extracts I would need to boil water to get water soluble compounds. With sonicator it is only possible to extract alcohol soluble compounds and I will need to make boiling after also to get water soluble compounds if I want double extraction? Or sonicator can not only make alcohol extraction but water extraction in cold water without boiling too?

Firstly, the sonicator operates at lower temperatures compared to the Soxhlet, minimizing the risk of thermal degradation of heat-sensitive compounds present in mushrooms. In Soxhlet extraction, the extracted compounds are exposed to heat for a long time in the heated flask. Secondly, the sonicator provides better control over the extraction process, allowing for precise adjustment of parameters such as amplitude, power, temperature, and duration to optimize extraction efficiency. Additionally, the sonicator has a higher capacity, enabling the processing of larger volumes of mushroom material in a shorter time frame. Contrary to the misconception, the sonicator is not limited to alcohol extraction alone. It is capable of extracting both alcohol-soluble and water-soluble compounds from mushrooms, including polysaccharides and other bioactive components. Furthermore, ultrasonic extraction in cold water is possible without the need for boiling, making it a versatile and efficient method for extracting a wide range of compounds from mushrooms. @DanielDeai Our antifreeze additive is pink. The antifreeze additive in the water reduces the temperature at which the water freezes to below zero degrees Celsius. This allows for a lower coolant temperature and better cooling. The pink color of this antifreeze is typically due to specific dye additives used by manufacturers to differentiate it from other types of antifreeze, such as blue or orange variants. Hi! First thanks for such a great video. I’m a high school student and I’m trying to extract from plants called Capillary wormwood and Prunella vulgaris L. subsp. asiatica (Nakai) H.Hara to see it’s Antimicrobial property on oral bacteria. But I’m not sure how long I’m supposed to do the Soxhlet extract and if I have to operate the Roto Vape to evaporate the ethanol. Also does it matter on how much ethanol I use? I am going to do the same amount on both of the plants but I wasn’t sure on how much to use. And another question.. Because I bought the plants and I don’t know what kind of things came with it, I’m going to make a batch washed and one that isn’t. But I don’t now what to wash with it. Should I try distilled water and tap water or anything else? Again thank you for a helpful video!!

The time for Soxhlet extraction depends on many factors, If you don't have any guidelines, we suggest you extract until you don't see any coloration of the solvent in the soxhlet well (compare to 4:15 in our video), meaning that there are no or few color compounds extracted anymore. To be safe, add the same time on top of that. You don't need a RotoVap. You could just boil of the ethanol - preferably into a condenser. The amount of ethanol does not matter much. However, the level in the flask should high enough to ensure filling and draining of the Soxhlet well, without running dry of overflowing. Instead of Soxhlet, you could just put the plant material in cold solvent and sonicate using a probe type sonicator, such as the UP100H. That will get the job done much faster 😉. Soxhlet extraction, while effective at extracting tomatine, is time-consuming and less energy-efficient. It typically requires several hours to days, operating at high temperatures with continuous solvent cycling, which can lead to the co-extraction of unwanted compounds, resulting in a crude extract. Sonication, in contrast, uses ultrasonic waves to disrupt plant cell walls, releasing the tomatine more quickly and with less heat exposure - at sub-zero temperatures, if you like. This makes it more time-efficient, reducing the extraction process to under an hour, while also preserving the integrity of thermally sensitive compounds. Additionally, sonication is more adaptable for large-scale industrial applications. High-power ultrasonic homogenizers, such as the UIP6000hdT allow for scalable batch or continuous flow processes, making it a superior choice in terms of overall efficiency and scalability. In terms of solvents, both ethanol and methanol are commonly used for tomatine extraction due to their ability to dissolve polar glycoalkaloids like tomatine. Methanol is particularly efficient for penetrating plant materials, but ethanol is often preferred in food and pharmaceutical applications due to its lower toxicity. Sonication combined with these solvents typically yields a finer, more selective extract compared to Soxhlet extraction, which often produces cruder extracts due to prolonged solvent exposure. The faster and more controlled nature of sonication reduces the risk of co-extracting undesirable compounds like fats and pigments, resulting in a purer tomatine extract. For both laboratory-scale and industrial-scale extractions, sonication, particularly with ethanol or methanol, offers significant advantages in terms of time, efficiency, scalability, and extract purity. Sir, I have some questions regarding Soxhlet extraction. I am performing a Soxhlet extraction using HPLC-grade methanol as the solvent and at a temperature of 65°C. The extraction proceeded, but after a while, it stopped, and the solvent stopped boiling as well, even though I maintained the temperature conditions. Could you please explain why this is happening?

Methanol should evaporate at 65degC. A) Even though you're maintaining the heat, if the cooling system (like the condenser) isn’t working properly or there’s a leak, the solvent could be lost. Double-check the cooling water flow and make sure there are no leaks in the setup. Also, confirm that the solvent level is adequate for continuous cycling. B) Your heating mantle or heating plate broke. We had this happen before, when we bought a cheap heating mantle. Had to buy another one and repeat the whole process. Cheap is not worth it ;-) Better use ultrasonic extraction at low temperatures instead of Soxhlet. Better extraction results, shorter extraction times, less cleaning and no hot methanol vapors. @John-of6ch No problem. For mushrooms, ultrasonic extraction is better suited than soxhlet. As explained in the video, in Soxhlet the extracted compounds are exposed to heat in the flask for a long time. That is not good for heat sensitive compounds, because it will cause heat degradation. So you are destroying what you just extracted. With probe sonication on the other hand, you can - break cell walls - Soxhlet does not, and - extract soluble and in soluble compounds - Soxhlet does not extract insoluble compounds Most importantly, ultrasonic probe extraction produces great yields and works at low temperatures, such as -20degC. @brandnewayurveda1835 How about using ultrasonic extraction instead. Soxhlet was invented in 1879, long before sonication. Sonication outperforms soxhlet easily. Just pre-chill your solvent in the a fridge or freezer and ultrasonically extract at low or ultra-low temperatures. This preserves heat-sensitive compounds, is much faster and more efficient. @momsmushroomsjodyfoster5786 Water can be used as a solvent in a Soxhlet extractor, but it will extract water soluble compounds only. Without sonication, the water or solvent will not break cell walls. Therefore, extraction is very slow and incomplete. @symptomesdelau-dela5442 The confusion about Soxhlet extraction being a cold extraction process comes from the fact that the condensed solvent is cold when it drips onto the sample, where it extracts the compounds from the sample. However, the overall process involves boiling the solvent, making exposing the extracted compounds to heat for a very long time. This heat exposure can be problematic for heat-sensitive compounds, which is why our ultrasonic probes are in high demand for botanical extraction. Our sonicators can operate at very cold temperatures, even down to 30 degrees below zero, preserving heat-sensitive compounds and ensuring high extraction yields and quality. @tparty22 The key difference between Soxhlet extraction and simply soaking and evaporating a sample in solvent multiple times is the automation of the extraction process. When you just soak a sample in solvent and evaporate it, you may not extract all the desired compounds, especially if they are present in low concentrations or are difficult to dissolve. You would need to repeat the process many times, which can be labor-intensive, inefficient, and may still not yield a complete extraction. Soxhlet extraction is designed to continuously cycle fresh, clean solvent over the sample. The solvent repeatedly passes through the sample, gradually dissolving the compounds of interest and ensuring they are effectively extracted. The advantage of this method is that it allows for a more thorough extraction without needing to replace the solvent manually or evaporate it multiple times. This makes Soxhlet extraction useful when working with large volumes of solid material or when you need to maximize the yield of extracted compounds. In summary, while both methods involve solvent and evaporation, Soxhlet extraction automates and optimizes the process, leading to a more efficient and complete extraction.
