A lot of media states her as transsexual, from a man to a woman, but if you really think about it maturely, and look into how she became to be, she is clearly female, but everyone in the Street Fighter Era say that she is as strong as a man. Ryu mentions that, also Chung Li. The creators said that she can be either or, which is stupid. So with that, I believe that she is a she that is extremely strong. Played with her whenever I can and she is really fun to play as.

However Capcom has said that in North American releases of their games, she's to be considered a post-op transwoman, rather than 'newhalf'. Of course they also say that part of the charm of the character is the gender ambiguity. After all, a lady has to keep some things secret, no?
Eh, I prefer to take the same stance as Capcom, and leave her ambiguous. A) because it maintains the interesting history behind her character, and B) she is a fictional character, so I'd like to think she's whatever to whoever.

It's officially stated that Poison is indeed a transgender. In america Poison is a post op trans, meaning she was once male whom got an operation to become female. In Japan she simply tucks away her business to look female. In my opinion I don't give a flying fuck if Poison is a male, female, transgender or whatever! She is hot, badass and i love her! Also i'm glad she is transgender because it makes her different and Street Fighter after all is about the diversity of fighters around the world.
In the original concept art of Poison, before final fight was even release, she was considered a new half which in Japan means a transsexual or transgender woman.
Poison and Hugo are based off real wrestlers. Poison is based off "Hollywood" Jeanne Basone from GLOW. Hugo is based off Andre the giant back when WWE was called WWF. Of course I know what Poison actually is, but it's not those years anymore. She's still strong and hot.

Poison was Originally female. When the game got ported to north america, there were concerns about americans having trouble accepting the idea of a woman getting beat up in a video game, so she got turned into a transvestite. But the americans still didn't like it so they swapped the character sprites altogether, but the transvestite idea stuck. So basically, it's region specific. If you're playing this in Asia, it's a female, if America, it's a male.
but seriously, ANY VIDEO where you have more than a minute of footage of Poison the comments become a whole gender argument! Similar to how its the same concept with videos' of Aeris's death scene. Its insane!

You're 100% correct, she was always transgendered. The difference is in Japan she still has male parts, in north america she got sugury. But the bottom line is that she started out male.
