Human error is not involved in chemistry analyzers. Both are equally accurate as long as the manual method is performed meticulously

Mouth pipetting of blood... What is this man. Don't do that.
@TheSingtangpaScienceGuy
@TheSingtangpaScienceGuy 3 года назад
Some clearly seem to be having doubts regarding this. And i'm starting to regret not having explained it in the video itself. The volume of blood to be aspirated into the RBC or WBC pipette is a miniscule amount compared to the overall size and carrying capacity of the pipettes. And on top of that, these pipettes along with their accompanying rubber tubes, are designed in such a way that there is practically no chance of the blood sample reaching up to even a fraction of the rubber tube, much less your mouth, even if you intentionally apply excessive suction force. Also, the end of the rubber tube that's supposed to go into your mouth actually has an additional tiny regulator valve with a capillary type hole, to prevent a rarest of rare accidental back suction. And on top of all this, i dont know if you are aware of it, but it is a well known and well accepted standard practice in clinical labs to use the mouth to suck the blood sample upto the 0.5 or whatever designated mark on the pipette when doing a count. To prove this, you can try sucking some pure water upto the 0.5 mark on the pipette and then intentionally try to force it up through the entire length of the pipette and the rubber tube, just to see if the water gets into your mouth. Hope that clarifies.

Thanks for the nice presentation. May I know, why I have to discard 1st 2/3 drops from Pipette, before CHARGING the chamber?
@TheSingtangpaScienceGuy
@TheSingtangpaScienceGuy 3 года назад
This is the same question i received from someone in the comments on my Video on Total WBC Count. Answer is the same. So i've copy pasted the answer i gave here. Hope it helps. "The pipette is designed in such a way that the liquid in the long tube of the pipette does not take part in the dilution and mixing inside the bulb. So to get the most accurate result, the liquid within this long tube (first few drops) is discarded. Also the liquid in the long tube contains only the diluting fluid. All the cells have been sucked into the bulb. So the diluting fluid in the long tube cannot not take part in the counting and so has to be discarded. Hope that answered ur query.

Why should we count RBC in smaller squares based on L rule?
@TheSingtangpaScienceGuy

Basically to avoid double counting. Heres a good link www.researchgate.net/post/During_cell_counting_in_a_hemocytometer_why_should_we_not_have_the_cells_touching_the_middle_line_at_bottom_and_right#:~:text=The%20point%20of%20this%20%22rule,between%20those%202%20squares%20twice.
