Can you achieve the same result using only 1 transfer matrix per interface, which includes the phase?

You can always combine two matrices into one by multiplying them, so multiplying the "boundary condition" matrix and the "propagation matrix" to make one matrix for each layer if you want.

Is the matrix at 22:19 the scattering matrix or the transfer matrix?

I only discuss the transfer matrix in these lectures. The transfer matrix and the scattering matrix are related, but they are not the same.
