The scene at 5:50 exactly mirrors what Nate went through with Sam in the beginning of uncharted 4. Also "you've got to let it go" could be nate trying to forget sams death.

This game was made 9 years before Uncharted 4. I don't think they were remotely thinking of Nate having a brother at this point in the series. It's just human decency to lift someone up when they're about to fall to their death

Tetanus, or lockjaw, is a bacterial infection caused by Clostridium tetani. Old houses, cars or other discarded items left in nature for long enough will rust (if they’re metal) and collect bacteria like Clostridium tetani. That's why Sully suggests Nate to get a tetanus shot or something.

Buka pintu means open (the) door in Indonesian/malay. The guy then says "who's there" then drake says "you idiot quick open the door" all in another language now drake obviously doesn't speak indonesian/malay. So that combined with the way he says it makes it a funny scene.
