If you are using your portable drive on both Windows and Mac OSX I personally recommend ExFAT. I believe it's slightly slower than NTFS but not by a lot and it also accepts huge files if you intend on back up bigger files than 4gb. ExFAT will read files larger than 4gb.
if you are using the drive for file transfers and storage, exFAT is just fine. But if you are using it for sample libraries (think KOMPLETE 14) or video production, it's usually better to have two drives - one for each platform, formatted in the native format (NTFS for Windows, APFS for Mac) support.native-instruments.com/hc/en-us/articles/360013984897-Storing-Libraries-on-an-External-Drive-for-both-Mac-and-Windows
