I came up with a good tek that doesn't need PC sterilisation. I use Glucose syrup which is osmotically bacteriostatic, sterile pee jars from the chemist and pure water from a cask which is pretty close to sterile and doesn't let air back in after dispensing. I use 1 or two colonised grains from a spawn bag kept in the fridge. Haven't tried with spores yet but so far no contamination. I keep my grain spawn in another sterile pee jar in the fridge

Mycelium can survive underground with very little oxygen. The amount of oxygen in the vial will be sufficient for the tiny amount of mycelium growing there, especially if it's constantly cooled

I've always thought it's the other way around (less nutrition = faster growth) since the mycelium is "searching" for nutrition therefore growing? I am confused.

Too much nutrition might be bad. Too little nutrition is not always good. The amount of nutrients should be optimal. Every species and even strain has a specific type of nutrients and their optimal concentration range.

Hello, my name is Sergey. Tell me, do you have experience storing mycelium on slanted agar in a freezer (-20 C)? My experience can be said to be 50/50. Half came to life within a week and the other half did not. I began to suspect that half of the mycelium was lost not due to freezing, but due to drying out. What experience do you have in this matter? The result is the same both on grain and on agar.

What happens if the honey caramelises in the pressure cooker. Is the LC still viable?

You should not allow any caramelization to occur. Such substrate is know to be unviable at least for most fungi.

Your best chance is that you will catch some spores. So go for the spore gills :)

I have a question, can we make liquid culture from dried mushrooms , using the inner tissue of the stem or the cap ?

No it's not possible. Dried tissue is dead tissue. If the sample tissue happens to contain spores - they will most likely germinate however.

We are pretty positive about that. Dry fungi matter is dead. But there is a chance that one could get some spores on dried fruiting bodies so it can still germinate. However the sterility is very much compromised in this case

Liquid culture solution (already viable mycelium) or liquid spore solution (spores will germinate and produce mycelium, spores are generally slower to colonize the liquid culture Medium)

It should be kept in dark place at the temperature suitable for spawning/incubation that corresponds to your fungi species.

Leave it in an area that is undisturbed, not too wet, you dont want any contam in the proximity of your Operation. Remeber that alot of contam is on the floor, alot airborne, always disinfect ur hands, any surfaces you are working on with Alcohol and any Equipment you use that directly Touches agar, liquid culture, spawn bag etc such as knifes and syringes should be Flame sterilized. Flame sterilization is best done with a "clean" Flame like from a butane torch. You can use a lighter to, the Black stuff "soot" thats left behind is completely sterile too so dont worry. Anything you flame sterilize should glow a bright Red, after Flame sterilization do NOT wipe down with alcohol, the thing you sterilized will go from sterile to sanitary (there is a slight difference)

Magnetic stirrer? Sure we know that. The question is, how would you benefit from using it... :)

Ah I see what you mean. Still though, we're not sure what magnetic stirrer would do. It might interfere with the development of the mycelium, or it might help it. My guess is it would make no difference. But of course, its all just guesswork until someone does an experiment with a control group to check it. It would need to be a monoculture grown on the same batch of nutritious liquid media.

I'd u put I. Fridge to lower temp it's gonna slow down growth a lot and can be kept for weeks or a couple months in not longer, as long as it's still looking good

Why did you put pressure into the saline bottle to let the water run out of another needle? Just stick the needle in, suck the saline into the syringe and use the syringe to place the water where needed. (????)

It's a basis for drugs that are administered via intravenous infusions in medicine. Saline solution can be used on its own for hydrating patients intravenously.

Yup, you just need to sterilize the substrate in a sufficiently large pot. The boiling should be for no longer than 20 minutes

100ml of water is about 100g. 4% is 4g. Why 1,5g of honey?

Having a chemical background we do know that a 100ml solution will not weight 100g. But it is close enough :) We found that particular proportion to be effective.

100ml H2O = 1801.528mol Honey (C6 H12 O6) = 180.1577g/mol 4% of 1801.528mol = 72.0611mol so we need 72.0611mol of honey which are about 0.4g. (keep in mind honey varies, so this is not 100% accurate)

You can use plain fructose (not so difficult to get). We haven't tried sugar but we heard that it's better to avoid. Honey is most preferable for it's high fructose content as well as micronutrients

Yes, you can use jaggery/raw sugar or even plain sugar (dextrose). However the growth rate (since dextrose takes longer time to break down compared to fructose) will be slower thus it will take longer for the mycelium to form.

Honey has antibacterial properties that help out. I’d stick with honey

We don't think that antibacterial properties of honey are strong enough to make any difference. Not in such concentrations of honey anyway. However honey is preferable because it's much more rich in nutrients than plain fructose, which has no micronutrients whatsoever.

Yes, You should keep your honey percentage in distilled/sterilised water from 3-4% not more than that. For example you are using 100ml of distilled/sterilised water then you have to use 3-4 gm of honey. If you use more than that then it can might destroy your mycelium.

Yes, 4g for every 100mg of water, using oat based mixure cuts the dwell time like (dried or ground oats/ light malted yeast or ultra light M.Y.) mean it will start colonizing faster, using sugar based like honey, could be a couple week before u see growth in the grain

Mushroom academy, you should post a video on how to make a contaminant free spore solution, then this video wouldn't seem like such horseshit. Creating a lc is the advanced thing that you're showing people how to do wrong. If you can't master agar, lc is out of your league.

@@trevorholliday7020 Actually LC is rather easier then agar, on our opinion. We used to do things a lot more strict in the past. Maybe we are just lucky but what we are doing actually works. But you are right, maybe we should make a video with a more strict adherence to aseptic practices.

Why ate people only using simple sugars only? Liquid culture like agar to thrive and not degenerate requires a starch source. Using you would add malt or some other starch source!!!

If you store it in the fridge, then it's good for at least 6 months. We have successfully used a liquid culture which was much older than 12 months.

We never tested the shelf life in room temperature. I guess it would vary depending on fungi species that you use, but at a room temperature anything beyond several weeks is a wild guess.

As i understand it, you have to use the liquid mycelium to make mushroom spawn, with either grain, wood fibers or coffee waste. Then you can start growing the shrooms when you have a strong spawn going.

It may work, way higher risk of contamination, longer grow time and less yields are to be expected Edit Would only ever try this with gourmet mushrooms, dont do this with medicinal mushrooms itll probably go to waste

Look for the odd mycelium colors (i.e. that are not natural for mushroom species you are growing). You can do a test run on agar.

Its hard to Spot contam compared to agar. Always test the liquid culture for contam on a few agar plates or you might Ruin a fine grain bag/ whatever ur using with contam
