Can we really cure ageing? David Sinclair thinks so, and he's going to try to prove it.
David Sinclair is a scientist and entrepreneur working on increasing human health, productivity, and lifespan. After co-discovering a molecular cause of aging at the Massachusetts Institute in Boston in the mid-1990s, he joined the faculty of Harvard Medical School where he is now a tenured Professor. In 2005, he founded the Glenn Laboratories for Aging Research at Harvard and serves as their Co-Director. He is also a Professor at the Lowy Cancer Centre at the University of New South Wales, Sydney.
His research is aimed at understanding why we grow old and using this knowledge to prevent and treat both rare and common diseases. He is perhaps best known for identifying resveratrol from red wine as an anti-aging molecule. He has founded four biotechnology companies; to treat age-related diseases (Sirtris), improve female reproductive health and IVF (OvaScience), treat type 2 diabetes (Cohbar), and develop vaccines against malaria, chlamydia, tuberculosis, pneumonia, and cancer (Genocea).
In the spirit of ideas worth spreading, TEDx is a program of local, self-organized events that bring people together to share a TED-like experience. At a TEDx event, TEDTalks video and live speakers combine to spark deep discussion and connection in a small group. These local, self-organized events are branded TEDx, where x = independently organized TED event. "Take care of your cells, and they will take care of you." I am convinced, after a life of paying attention to my health, that we age and are healthy at the cellular level - or not. If we aren't, we have not treated our cells right. Looking at people who mistreated their bodies - most of them knowingly doing so - it is sad to see the cellular condition of their skin. And one can imagine what they've done internally, too. The approach of microbiologists is correct, IMHO, to look to the cells and their components to determine what is going on. They are making enough progress that it appears completely possible that they will some day be able to stop and reverse aging. I applaud their work and effort and encourage them to keep on trying to discover more and more. Maybe within 25 years, when I am 90, they will be able to take me back to 40 or so. I will be content with my 120 years or so, regardless. But if I can get another 20 or 50 or 100 years, I am all for it. There is so much to learn in life, and it seems such a waste to learn it all and then simply wither away. It would even be BETTER if there are a lot of other super-centenarians to accompany me. I believe in people, and it is people like David Sinclair that I believe in most. It may or may not be HIM who gets us there, but one of them will, someday. Break a leg, folks!

Am an avid fan of David Sinclair. I believe in his science and will try to reverse my age of 72,witj a heart disease and pacemake, a type 2 diabetic, meniers disease, vertigo, but not giving up.. Now I feel 40 years old. Thank you

I used to run the MIT track as a little girl. There is a diet called the MIND diet. It's a Mediterranean diet based on plants legumes whole grains wine and fish. Mostly plants real vegetables and fruits. That makes sense because there was once a saying -"let food be thy medicine"

Stunning! My own observational research postulated that eating too much works your cells out, making them age quickly, so by eating moderate they work moderate and by starving you switch of the division of cells. Now these increases the lifespan of the cells and whole organ. hahaha so impressive!

I’m 51 years old and going to be 52 in a week and I was seriously healthy all of my life until I had someone hit me at 60 miles an hour t-bones me I was knocked unconscious and I have struggled with pain daily from sciatic nerve damage until this very day the only injury I have ever sustained in my life time, I’m blessed to have had such great health and what did I do but started taking pain pills for the pain and became addicted, this went on for years from age 26 until 47 so I’m pretty sure it’s affected my liver I have not been diagnosed but it’s a feeling anyway my point to all of this is that I agree with the aging reversal because by eating healthy you are more alive so everything is working properly and you won’t age much at all. I know because I’m 52 this week and can take pix with 20 year olds and no one can tell they are not my girlfriends but friends of my sons lol I am just now starting to age and it’s been since being married 7 years ago, my life has seen times that no one should ever have to endure and I still look very young compared to others my age at least from what I can see anyway and I’m now working on my inside my self my attitude because I got the healthy part right thanks to my father I’m from the country and ate right out of the garden right from the ground washed it off and ate it! It works

I first heard Dr Sinclair in a radio program last year. I had already been missing breakfast for literally decades, but still had stabilised at around 86kg, down from around 100kg with 'dieting'. I began his recommended daily time-restricted eating on a roughly 18/6 regime, and 18 months later I'm stable at 65kg and never felt better! But I do have a lot if spare skin, if anyone needs some...

I just got David's book and it is amazing, I have not been so excited to read a book in so long! I have just started taking NMN and Reservatrol supplements, about 6 weeks now and wow I feel amazing! I feel younger, I have more energy, I don't get exhausted/tired as easily! Over the last 10 years I have been working towards being healthier, working out, eating right, and now with Davids research and many other scientists as well - this is an exciting time to be alive!

I am a moderationist eater. I eat meat or eggs most days of my life. I don't smoke, and my alcohol intakes are minimal to moderate. I am 65 and I look like I am 48 or so, according to new people I meet (and I meet quite a few new people in my life). NO wrinkles, anywhere on my body or face. Natural brown hair (what hair I have). Perhaps I have good genes, you might surmise. No. My two brothers (17 months older and younger than me) had premature grey hair and the none surviving one looks to be about 80. My older sister had quintuple bypass surgery when she was younger than I am now. I visited the Mediterranean in my early 20s and discovered what is now called the Mediterranean diet. I have basically followed that ever since, with a Snickers bar or a Coke every few weeks or so. Like I said, "moderationist". The diet I discovered is simply olive oil, garlic, onions, fresh produce, wine (especially red wines), cheeses, seafood, and lamb. To that I added eggs (about 7-10 per week, plus real mayonnaise), beef, chicken, goat, and real butter, all in normal quantities and all as un-tainted by chemicals as possible (and that is certainly not always possible). I also decided at age 20 - as an experiment in mental suggestion - to live another 100 years, to age 120. I am now 45 years into that 100 years, and I am healthier than any 65-year-old I know. Essentially all of them are wrinkled and white or grey haired, with withering bodies and slowing mental faculties. I have a belly that comes and goes, and I don't stress over a few extra pounds. I recently was buoyed over that, even, when a paper came out that stated that overweight people actually survive illnesses on average slightly better than people of the so-called ideal weight. I wonder sometimes if, when I projected myself to age 120, at age 20 I was somehow tuning into the research then beginning on aging and longevity. It seems like the world is trying to make me able to reach that 120 age. I am no soft-in-the-head New Ager, though I spent some time there. I had a career as a hard-nosed mechanical engineer, having worked my way up through the ranks. I assure you that in the world of heavy industry and very practical foremen and machine builders and welders and such one does not go through life with one's head in the clouds. But I know that I set myself an experimental goal 45 years ago, and it looks like not only that MY body is trying to get me there, but that science is also trying to get me there, too, carried along by efforts of people like David Sinclair, who is trying to save the whole world, me included. A year ago I bought some Reseveratrol for a girlfriend, but then we broke up before I could get it to her. It sat on my shelf for a year and now I am discovering some of its properties. So now I am trying it. I have NEVER taken pills for anything, but since it is made from red grape skins, I am giving it a try, just for the hell of it. I haven't gotten a buzz yet, but will report if I do. I actually offer myself to him and like-minded researchers, if they want to see someone who is aging more slowly and include me in their research. I am doing something right, and perhaps they can learn from me.

Steve Garcia I think it's a good idea to decide how long we want to live. I'm pretty sure it can have quite an impact on our subconscious level. Especially, when I see other people who are complaining all the time about aging as if at 50 it was impossible to learn anything new! I think we should rather look at people who run marathon at 100 and wonder why not us than getting stuck with stupid beliefs about aging.

We think of the Mediterranean diet as healthy but that's because the original Mediterranean diet was like 90%+ plant based before these people (in the last 1/2 century?) started eating unhealthier and now we think mediterranean = oil, wine, fish... NOPE. Steve Garcia .Hi steve...I.m in a similiar sitution to you..age 63.but look 45.full head of hair and sport a modest 6 pac.i eat mostly wild game .in small amounts and a plant full diet.i follow practices of blue zone places..exerise in bursts.and take many spplements.although i know how to grow food naturally if i didnt travel so much.ie garden etc.i adopted many foods most never heard of. ie.natto.moringa.bitter melons.and on and on..hope your in good stead.

Old news. There's a lot of peer-review on pubmed that actually demonstrates that Vitamin D deficiency heavily contributes to age-related diseases (especially on a mental level - cognitive decline, bone issues, immune problems, muscular problems, etc.) Keeping people healthy in their 'old age' isn't really difficult with what we know without synthetic drugs. A plant-based diet (vegan) also has peer-review data correlating overall cardiovascular health for people in their 90-ies... coupled with exercise, etc. The RDA for Vitamin D was miscalculated to the point where it ended up 10x lower compared to what it should be - an article was published earlier this year on Sciencedaily and PubMed. The RDA for Vitamin D3 should be in the range of 10 000 IU DAILY (for some people, it would be double - depending on their age, weight, etc.) What I'm more interested in extending the Human lifespan beyond the 'paltry' potential 150 years of age. But then again, Humans might not have a fixed lifespan. The oldest known person died at the age of 122... and these numbers seem to be rising. Peer-review on pubmed also seems to correlate longevity with high glutathione levels in the body of people who live past 100 and 120. Intracellular glutathione levels can be raised via supplements using Alpha Lipoic Acid... though R-Alpha Lipoic Acid was stated to be more potent. On an overall basis, NAC seems to have the ability to restore glutathione levels in people in just 2 or 3 months at doses of about 6 to 9 grams per day. Combining that with Vitamin D3 - at least 10 000 IU daily (I do 20 000 IU daily) and curcumine inclusion in vegan diet with regular exercise should be ok. Vegan diet also according to peer-review can easily provide your body with everything it needs in abundance (B12 included - Nori/seaweed). Most/all minerals can be sourced from almonds, brazil nuts almonds, flaxseed, microalgae oil and chia seeds. No need to chow down on these all the time though... just include them to be part of your regular diet at least 3 to 4 times a week. Out of most supplements, Vitamin D3 remains one of the cheapest ones... so if NAC is not an option for you, you can improve your glutathione via healthy vegan diet rich in fruits and vegetables. Combine it with Vitamin D3 supplementation, and other food stuffs included, and see if it makes a difference for you. As for extending life beyond the paltry potential 150... could conceivably be done today via stem-cells alone. It could be doubled... which means youth, vibrance and health (physical and mental) at an age of 500 (probably well beyond that) - well, basic principles apply in terms of body maintenance, so you will simply need to be careful that you ingest the things that are beneficial to your body. Also, by 2030 or 2035, nanorobots will enter the picture which in combination with regenerative medicine (in the next 5 to 10 years) will likely enable biological immortality.

+IVXCLD Already had my D levels tested. Was stated to be 91 ng/ml (227 nmol/L). As for NAC causing people to froth at the mouth or talking in tongues... hardly. NAC is a fairly harmless substance. I would imagine however that older people would need to be using smaller doses of NAC initially before increasing it to 6 to 9 grams per day (like anything, your body needs time to adjust) and benefit even more from Vitamin D3 supplements in doses of 10 000 IU to 20 000 IU per day, as well adjustments in their overall diet to shift them more to a plant based one. +IVXCLD 91 ng may seem high, but according to peer review, its perfectly fine. No problems were detected in Humans in ranges of levels under 250 ng (which in itself is a much higher number). As for minerals like magnesium... that's easily sourced from nuts (which are far superior when it comes to magnesium quantities compared to anything else you might be ingesting). Western diet has a high emphasis on meat, eggs and dairy - mainly because those industries are excessively dominant... therefore, it is very likely that plant based diets will receive a negative backlash when vested interests are put into question - even from peer-review. As for Aubrey De Gray... I agree with his idea on completely preventing people from ageing... but there is another way to achieve his 'escape route' which would slow down biological ageing to the point of proverbial crawling if not possibly even stop it almost completely while also preserving health and vibrance on a physical and mental level ... and some of those methods I already mentioned (they are all well within our current capabilities, but my point is, we don't need drugs to do it). Right now we still don't know very long-term effects of vegan diets on elderly populations. The only peer-review I'm aware of in this area is the one that established protective cardiovascular health effects on people (going beyond even what vegetarians achieved). For the moment, we seem to continuously increasing the maximum life expectancy (but those also missed other key factors that might have allowed these people to live even longer). I don't concern myself with 'average life expectancy' because that's just rubbish number which is quite frankly set too low and also cannot be applied to EVERYONE.

Well, keep in mind that vegan and vegetarian diets are high in protein as well. Peer-review demonstrated to my knowledge that the amount of ingested protein from vegan and vegetarian diets was effectively no less compared to those on an omnivore diet. Longevity might be correlated with dietary choices (considering that specific diets obviously have an effect on cardiovascular health, etc.) But it also depends on balancing what you eat, with exercise, etc. - so its 1 factor out of several others (genes play a very small role here though when you take into account epigenetics). So, it doesn't really matter what kind of background you come from, so long as you adjust your style of living and environment to one that benefits you in terms of longevity and health (I would imagine some of these might be negated by the western life which can be full of stress, but meditation in turn could help in this manner). There were plenty of people born in 1950... as for how many of them will live to 150 remains to be seen. Keep in mind that most of these people probably hadn't read into peer-review research involving longevity, etc., so most might not know how to go about it... or even that they COULD affect anything about it, because we live in a world that demands of a person to have 'money' to gain access to anything (which is something society needs to change dramatically, because that kind of outmoded notion of societal conduct has no place in the 21st century). Enache a) I have had no backlashes from using 20 000 IU of Vitamin D3 daily. b) I have been taking Vit D3, between 10 000 IU/20 000 IU for almost 1 year and 10 months now c) In terms of visual evidence, I noticed that my skin is no longer dry for example and is much smoother (prior to starting D3 supplementation, I had dry skin which was relatively rough as well - this does correlate with peer review findings that D3 affects innate and adaptive immunity). d) I am currently taking NA-RALA (a supposedly better form of R-ALA) occasionally - however, glutathione can be regenerated through sensible diet as well. e) Been taking NA-RALA on and off for the past year f) Nothing in terms of visual evidence, but considering that my chronological age is 32 (whereas my biological age seems to be placed at about 10 years below that), I am vegan and also go to the gym 4 times a week, I would imagine that I wouldn't notice too many effects as opposed to someone in their 50-ies - though it is possible that due to younger age and these precautions, I could be slowing down my ageing (or affecting my genetic structure in certain ways) for the better without even knowing it. I doubt that I might be doing my body harm though, since the doses I'm using don't seem to have any harmful effects, and been using them on a fairly long term basis now. I don't have my own blog really or a website. I just like spending free time researching peer-review and experimenting with supplementation towards life extension, etc. and try to base my replies around peer-review as a basis.

I'm 19 and I truly hope that there is a cure for aging in my generation. I'm a nursing student and I see the effects of aging daily. If we spent more money to fund aging research, it would do much more in preventing the diseases that we are trying to cure.

I totally agree, Sameer. I can't believe people would just choose to die at the age of 100, sick and elderly - and I can't believe people just cast aside longevity research with the statement that "death comes eventually for everyone". You can activate those SIR-2 genes or "pacman" through exercise and fasting. pharmaceutical companies know it's more important to keep people sick or dependent on treatment than when we are healthy and with slowed down aging processes.

I would love to see a lavishly funded international life extension/immortality research and development program similar to the Apollo Program. We do not know what we are capable of. At the very least, my guess is that we can likely extend both the average and maximum healthy human lifespan, and that would be good.

I will be 85 by the time the FIRST things will be experimentally tested! And that is still decades from freely available cures! Get OUT while you can! Life is NOT meant to be lived until high age. +im going on holiday to Vietnam date Research has discovered that the life-extending aspect aspect of fasting is methionine -reduction. For the most part, this is accomplished with the diet I posted above. There are also many plant foods high in methionine, (Brazil nuts the extreme example), so even plant-based foods must be chosen carefully. Obviously, overeating would still have a negative effect, even on this diet, but these foods have high water & fiber content, & no man-made appetite-stimulators, making overeating unlikely.

Unfortunately, while those certainly do help, there are things you can't stop such as oxidative stress. NAD+, Pterostilbene and SIRT1 activation can help with these things.

People who say they don't want to live forever should still support this because you can always change your mind later but not if you're already dead. I have been saying this... longevity is our choice! If we want to sustain our health, we have to do something about it! This will start by knowing what supplies our body needs. Start by eating a balanced diet, quit vices, exercise regularly and take an NMN supplement. Mine is a 500mg per day, NMN by Vitruvin Nutrition. The goal should be to live longer while staying young and healthy. Sinclair believes that in the foreseeable future, it's possible for humans to live beyond 120 or even 150, without the frailty, diseases and disability we now associate with old age.
