( They were originally made of brass because brass vibrates, very important when throwing a punch ) basically it is a weapon meant to increase the severity/lethality of a punch. Even if you had a weak punch brass knuckles will leave a mark, they are used mainly in street fights and it is illegal to use in competitive fights like boxing or mixed martial arts. But on the streets its legal, its just meant to protect you while giving you a better harder punch.

Also Batman immediately recognized it as synthetic so it is possible that he used that as well like in the brass knuckles. Though I don't doubt that Batman has real Kryptonite but it's not like he wants to actually kill Superman.

I trusted him with my life. But then The Joker got to him. I lost my friend, Clark. I've missed him ever since." And there's also the "Death of Superman" plot in Justice League where Superman is seemingly killed by Toyman, and Bruce is the only one who refuses to believe that Clark is actually dead (and Bruce is *right* to boot).
