Can anyone tell me why we specifically need 72°C we can just lower the temperature and use DNA POLY of normal bacteria instead of any thermophilus bacteria

I went through the video. Its quite informative but again I have few queries. I would be glad to get response. Queries are: 1) Is it possible to get replica of whole content of human DNA as its length is n= 3300000000 bp? 2) What are these primers? If they are RNA primers then, there must be Uracil (U) in place of Thymine(T). How it is again converted into thymine as there must be no 'U' in amplified DNA. 3) What is the size of primers and from which organism it is isolated, as it is very hard to isolate RNA (being unstable)?
Ans 1 As mam told pcr can be used for short segment of DNA. I think 10kbp max. Primer is removed after DNA synthesis by an enzyme I think primase (not sure) I think primer size is about 15_17 bases long. It can be synthesised artificially for pcr. In cells they are derived from RNA
Mam ,. Thank u for all ur videos on science , mam please if possible , can u make videos on how to make buffers and how to solve the pH of that particular buffers , because it's most important in so many experiments , it is useful for students who are doing research..thank u mam
Mam, you rise the excitement in learning every day mam .......on top mam .....no words about your teaching mam .Because of you biology made my favourite of all....👌
