Yes and there are multiple barriers to P.O. drug administration including: resistance to stomach pH, digestive enzymes, absorption, also the liver's ability to metabolize the drug. All of these would go into the dosage consideration. The effect of the liver on the G.I. absorbed drug is called first pass metabolism. Some drugs are activated when they are metabolized by the liver.
