I first made the backing track with Band-In-A-Box and played the melody along on harmonica whithout recording. Then muted the bass part and recorded with the bass harmonica. At last I added the melody. I was supposed to end up having only one sound track, but the bass was to weak and I then added the clean bass sound from the camera recording. I was not meant to be that complicatet! I record the sound via a good microphone and a mixer using headphones to have the best sound possible - and I take the video with a Logitech web camera that also happens to pick up the sound from the room. Normally I just use that soundtrack to syncronize video and mixer sound.
