In this video of DNA replication we have discussed about the enzymes involved in the Prokaryotes and Eukaryotes.
n DNA Replication , the Single DNA molecules having two strands is replicated to form two DNA molecules…. we see we having DNA molecule,its two strands acts as Parent strands to make two DNA copies ,so on these parent strands the enzymes and proteins will work on to synthesises the daughter strands on both the strands ,so that in the end we get two identical DNA molecules.

Pol €( pol epsilon) - it is recruited to complete DNA synthesis on the leading strand. It is associated with Proliferating Cell Nuclear Antigen(PCNA) protein that serves as a sliding DNA clamp.

thank you very much for DNA replication series. Very informative, very accessible form in which you've presented such a hard topic for me. Can you answer a couple of questions, please? 1. As one of the commentators has already said that eukaryotes primase activity is built in alpha-polymerase, can you clear the telomerase replication plz? during this, the primase is also a subuinit of the alpha-polymerase or it is some kind of special one? 2. As far I can understand eukaryotes have a clamp loader and sliding clamp which are independent from delta- or alpha-polymerase and prokaryotes have gamma and beta subunits of polymerase III for that. Is that correct? 3. I understand that it might be a complicated one, but I don't grasp whether there is any difference in the unwinding of dna (during initiation) between euka- and prokaryotes. For the exception of that pro's have gyrase and euka have topoisomerases. I mean the negative coilng part and chain stabilization in eukaryotes (not that they have orc instead of protein A). You have a done a great job, appreciate it very much Sir.

1.Yes Dna Polymerase Alpha has built-in primase in Eukaryotes PRIM1 and PRIM2. 2.Ur 2nd statement is right. 3rd statement is complex one and i need to see it how it drives... Btw thanks for appreciation..Glad to know that it helps...😊😊😊

E (eta) polymerase synthesises leading strand, d (delta) polymerases synthesises lagging strand. mentioned in watson's molecular biology of the gene.

Hi sir may I know for DNA repair, the enzyme that involved in proofreading is DNA polymerase III for prokaryotes right??? Then the enzyme used to replace the damaged nucleotides is DNA polymerase 2?? Or both proofreading and replacing the nucleotides are done by DNA polymerase 2 ????

Polymerase 3 has exonuclease activity once it completes the replication from 5 to 3 prime then it go for 3 to 5 prime exonuclease activity(proofreading ). After this in case still any wrong nucleotides are present on dna strand then DNA polymerase 1 will come and repair it. Dna polymerase 2 also involve in proofreading activity or dna repaing activity but correct function is still unknown.

DNA polymerase moves along the template strand in a 3'-5' direction, and the daughter strand is formed in a 5'-3' direction.

(1) Our observations indicating the prevalence of L612M Polδ generated signature mutations on both the DNA strands in pol3-L612M msh2Δ at different positions in the genome in two different S. cerevisiae strains provide positive proof for the conclusion that Polδ replicates both the leading and lagging DNA strands. (2) In agreement with the role of Polδ in the replication of both DNA strands, genetic analyses with the pol2-M644G Polε mutant lacking MMR have failed to provide any evidence for the involvement of Polε in the synthesis of the leading strand. (3) We provide evidence that in addition to MMR, Polε exonuclease and Exo1 function in the removal of Polδ replication errors from the two DNA strands, and that these different mismatch removal processes can act differentially on the leading and lagging DNA strands. We suggest that yeast strains differ in the relative contributions of different mismatch removal processes for correcting Polδ errors from the two DNA strands. (4) Previously, it was concluded that in S. pombe, Polδ and Polε replicate the lagging and leading strands, respectively. A reconsideration of published data, however, implicates a role of Polδ in the replication of both the leading and lagging DNA strands in S. pombe also.
