The Enclave didn't create the mutations. They found a way to make serums that induce specific ones intentionally, as opposed to the random ones earned through radiation exposure. Lore wise, they wanted super soldiers. Gameplay wise, they allow you to get specific mutations that will benefit your build without having to get them randomly. Fun fact: even if you have the mutation a serum provides, drinking it will still temporarily nullify a mutation's debuff.
The Circus is a hub for events, while it does hold some interesting characters to talk to at times, most of the time you come here to do events. the reason why it feels so empty is mostly because of that. Hopefully you catch onto the public events to see how goofy this game can be.
Btw Nuka world on tour its a place to do group events that why not so much to do there. Just unlucky time no event was going at that place plus these events may count toward Modus quest.
the events themselves don't count, however there are guaranteed legendary enemy spawns in 'Most wanted' (sentry bot), 'Tunnel of love' (2x Deathclaws), 'Seismic Activity' (ultracite Titan) and possible legendary spawns in 'Spin the wheel' all of which count for the quest.
I do enjoy listening to MODUS speak. It's a bit of a shame that they haven't done anything else with him, or almost any of the NPCs from the base game aside from Rose, in any of the updates.
