Bear in mind that the first time we hear the song Crackerjack and Honey Sugarman are singing it together, and it's joyful - of course, Joseph (boo hiss) cuts them off after the first A section with the line about "time's arrow," so we don't get the tragic lyrics of the following sections ("thoughts of you haunt my heart," "I don't want to be alone now," "I keep trying to escape this town," etc.). When we hear the full song with Honey and Eddie's duet across time, both singers are expressing their grief, which is all the more poignant because we know that they don't have a duet partner in their own time lines: we know that Honey and Crackerjack used to sing it, and it's implicit that Eddie sang it with his now dead wife. We then get three-quarters of it on solo trumpet in "The View From Halfway Down," which is a harrowing context. We tend to associate the song with tragedy because of the duet across timelines (the only complete performance in the series), which is one of the most memorable scenes in modern television, but the song within the world of BoJack Horseman probably was sung more frequently like it is here.

I'd also tie it to the Tin Pan Alley tradition, as it's essentially a 32-bar AABA (it totals 35 bars due to two introductory measures and then an extra measure at the end, the latter essentially a written-out ritardando).
