The simplest estimate of IC50 is to plot x-y and fit the data with a straight line (linear regression). IC50 value is then estimated using the fitted line, i.e., Y = a * X + b, IC50 = (0.5 - b)/a.

Is this accurate? Because when I plop this data into a tool for calculating IC50 I get vastly different results.

May I know if we set the graph with set intercept, what about the R value. Is it okay if it is not nearer to 1 ?

It's the standard deviation from the mean. So for the first concentration, it's saying the mean cell viability is 86.6% +/- 7%. Not sure if I've explained it very well 😅

From dose response curve and calculating the x from the curve, but make sure your R2 value is above 0.95 for good interpretation.
