I’m the last person to give you advice. Your knowledge on so many subjects is truly amazing and if I had just one percentage of your knowledge I’d be transformed. But, it strikes me as odd that something that grows naturally in the wild should be so fragile and hard to grow in a purpose build shed. Could it even be that your environment is just too sterile and if other microbes were allowed to just propagate perhaps some kind of natural balance could be achieved. Even from a consumer point of view, I’d want to buy a product that grew as close as possible to nature. I get the same vibe from hydroponics in that they use science to provide supposedly optimum nutrition and yet there is just something about it that leaves me cold. Of course it would still be preferable to growing in heavily polluted soil. On the teeth, I have also suffered from bad gums for years. More recently I stopped using fluoride toothpaste with all the nasty chemicals it contains and switched to a herbal one which seems to have helped. I also got both glove oil and actual cloves and lately I chew on a couple of cloves twice a day. Cloves are known for not only relieving pain but apparently they can also inhibit certain germs that might be contributing to your condition. Diet probably also plays a part and you might be able to single out certain foods that don’ t agree with your gums. I am also in a situation where I currently have no idea how I’m going to find the money for next month’s mortgage payment so I’m unfortunately not in a position to give any financial help. Dave you are so kind. Thank you. and re the teeth- Its been a whole year with ulcers-3 different prescription antiniotics that did nothing. Now on MMS and DMSO mouth wash which seems to keep it at bay but the gum recession is tragic!
@@t.h.o.r. wishing you all the best. Teeth, I only have my own experience ( supported by a ton of similar stories), My teeth were moving around, the gums were that bad. I stopped eating plants, and the gums healed, holding on to whats left of my teeth.

Ok, I am not a mycologist but I have worked with microbiological cultures for a few years in laboratory condition, so take my suggestions with a pinch of salt. From the information you provide it seems that you are very much overcomplicating all of your procedures. You are introducing many points of failure and possibilities for contamination. I have worked in labs ranging from the simple benchtop run-of-the-mill school-type setups to the environmentally controlled labs with positive-pressure cleanrooms. Your setup compared to my experience needs to be simplified. Remove as much technology as possible and culture from basics (sell to get some compensation). If there is still an issue then add one technology and see if things improve. If there is still the SAME contamination, try another single technology. In terms of issues like condensation on plates : do you turn the plates upside-down after the agar has just set? This causes the the moisture to 'rise up' into the bottom of the petri-dish. I think for people (including myself) to correctly help you with advice, we need to actually see you do the procedures, so we can see exactly how you execute your protocols. Are you using correct aseptic techniques when pouring the plates : barely lifting the lids of the dish, moving it half a centimetre to the side to pour the agarose gel slowly to prevent air being drawn in, for example. You use multiple reagents and other elements to create the cultured plates. Have you tested each item individually by plating them up and seeing if anything you are using is causing the contamination? We need to see your SOPs and also the recipes for your plates and reagents. Maybe you need to adjust the recipes in some way. There is so much to cover but that provides hope in finding a solution. As for the effort it would take, the investment you have already sunk into the venture makes it important to try whatever you can. I need to be honest and please do not take this as an insult but there is a certain level of foundational skill and experience that leaves many self-taught people at a huge disadvantage. You would benefit from downloading a simple lab protocol textbook and going through the basics, while also contacting a local university to ask if it is possible to do a bit of free work experience for a week perhaps. Sorry I that I have not been that helpful. I just don't have enough information to be able to make better suggestions.
aww- come on! not that helpful? you are awesome. and so kind with your time. 😃 so much of what you say had me nodding, checking things off. You are dead right. Im a fabrication guy and mechanic who is learning how to be self sustaining. Not a classically trained microbiologist . Im always up for a challenge though, so I jumped in deep- to learn it all. The mushrooms will help feed my little family and give us an income. Ive had no real income this year at all. Its just a savvy lifestyle that is keeping us afloat So its a bit grim right now. The shrooms will be a game changer for us as I cant work anymore with bung shoulders. I should be simply buying in ready to fruit blocks and just get the final part of the process perfected while I work on my contamination without stress. but for it to be worthwhile I need 10 blocks per week minimum plus $10 each freight till I sort this problem- trouble is- I dont even have enough money to fund that. Its a tide changing situation If I stop that contam- Ive got $3000 per week coming in from each crop.
Oh- and I was confident that if I can build cantelever suspension setups on a single seater race car - surely I can grow mushrooms! There are several 'mushroom Teks' that different size growers use from shoe boxes and wood shavings to glass jars and coffee grounds. Because commercializing this is a priority, I decided to start with what I wanted to end up with and built a small commercial farm to fit the building that I have available. Yes there is a bit of tech- but that all works as it should. Its just the evil contam in my triple sterilised dishes!

Hi mate, regarding condensation on agar plates while they cool, try this: 1. pour, and immediately cap, 2. wrap the dishes with micropore tape to seal the dishes lid on. 3. once cooled to 'not runny' state, turn the plates up-side-down so condensation drips onto the lid and not the agar. Humidity shouldn't be an issue, Mushies love humidity If it always seems to be in agar, try using a liquid culture. I've had my best results with liquid cultures
I was thinking this as well regarding liquid culture. I never tried agar plates but was planning to make liquid cultures. I suppose you have watched a lot of mushroom growing channels right? I bet some of them could get back to you if you reach out. I wish I could help more but I've only used growboxes but once when I ordered from another vendor the mushrooms got mold, either it could be something they did wrong at the plant they prepaired them or it was because I later bought a heating mat to use below which made condensation to appear.
A STILL AIR BOX might be your saving Grace, cheapest solution for highly contaminated work spaces. You make it out of a clear storage buckets just 2 holes for your hands you spray it down from the inside. The Agar plates keep them UPSIDE DOWN after inoculation.
Im 64 and my body is broken and I work 12 hours per day 7 days a week. - thats why Im getting into growing- because I will not ever be on a government benefit! just saying!
OKAY MATE , You want to create sterilized Agar plates and you your leave them for 1 hour in Each Location you work. You seal them up and you will see what contamination is in that Area. You can Also put Agar plates in your house. You make have to work in your house instead. Try using a Still AIR BOX you can make them out of clear plastic Tubs. As far as the moisture in the Agar plates , you can keep the Agar UPSIDE DOWN after inoculation. You want to keep the water pooling on the lid not the agar
Yes- but its mainly for water- uv lamps to sterilise a room would cost a fortune- Im gobsmacked that the Ozone and Hydrogen peroxide and white vinegar are not doing the job! I huff Peroxide through the HEPA filters before doing any flow hood work and 3 days later....... contamination on every dish!
It's amazing mushrooms grow in the wild if they are so finicky about cleanliness .A friend just has a pile of tree trunks in the back of his large greenhouse and grows some incredible chicken of the woods mushrooms . Maybe your grow room is too clean and friendly bacteria is not keeping the nasty bacteria at bay . You are pissing in the wind trying to eliminate bacteria even with the best filters ect and you need to let nature help you out . I think the nasty bacteria is only surviving in your set up . I have seen some great mushroom production just using boiled straw as bedding . I think your over thinking the problem and your sanitation is selectively breeding the nasty bacteria.
Hmmmmm? now that is a whole new concept. Ive watched hundreds of hours of awesome videos from successful growers and nobody is suggesting your approach. It appears that none of the 'friendly bacteria' got the invite. lol. Ive not seen mention of freindly or beneficial bacteria anywhere. Everyone is just going down the 'sterile path' - and it works! @@t.h.o.r. some experience with fungus here, and i agree, it's always suggested to be as sterile as possible. i have never heard info regarding some "beneficial" organisms that you don't want to destroy. pretty sure the idea is that nothing should be growing but mycelium...
Not sure if you've tried it yet, but perhaps look into using hydrogen peroxide in the medium itself to give more antibacterial defense. Peter McCoy has a book called Radical Mycology that covers thus process in detail.

I had a couple of thoughts. Is there any wild mushrooms that grow there? They do grow pretty well outside in their native areas. Maybe it’s just too clean in your lab which makes the conditions less likely for them to produce. Also, maybe it’s your product that you currently use might be contaminated. Sometimes as humans we tend to forget that mushrooms almost all the time like it outside with the right amount of humidity.
Yes we have allsorts growing in the forests and parks around me. Im providing a nutrient rich substrate and a perfect environment - so all the bacteria want to come and party! the mycillium is more fragile than competing bacteria and cant outpace it or grow over it. The whole idea of sterile workspace is to eliminate competition. and thus provide a regular quality harvest.
thats what Im putting it down to- But I did three waterblasts and disinfectant washes before I insulated and lined it- now I wipe down weekly with vinegar. It 'seems' my issue mat be with using Methalated Spirits to sanitise- Im now switching to Isopropyl alcohol - watch the future vids for my update in a week or two!

I think a comment that came in last night might be the key-Eliminate the Agar contamination by eliminating the exposure of agar to atmosphere- There is another way- liquid spores injected into the grain- less chance of contam. Ill order some spawn syringes and try this other method. Thank you for your offer of crypto- There is of course a cashflow problm- but that is secondary.
I had two people give me good suggestions which I am actioning now. "ditch the meths and use Isopropyl" and "stop using agar dishes and use liquid agar in a syringe- zero air contamination" so Ill know in about two weeks
Hello I once ran a northern Canadian year round grow op started in a simple seacan and grew to a major business before our farm was shut down due to resisting the CONVID agenda in Canada. So I'm basically back to small scale and from what I seen here your filtration is not sufficient. I tried similar to yourself with a 3 stage HEPA filtration system and it failed. I upgraded to laminar flow hood system at first a small 1'x2' single stage laminar in a pressure box on a simple table. That one change allowed me to be much MUCH more flexible on the rest of my procedures. And so because of this hard lesson that did cost me a few months I am convinced most branded filters that claim HEPA status ARE NOT. the laminar flow hood HEPA filtration if the only one that's ever produced consistent results while I've tried changing variable on all aspects before and after on my own hand pours petris and spore sources the most consistent fix is proper filtration. The grow rooms I only used a Merv 8 setup and had minimal problems as well. Temp and humidity were usually the deciding factors after a clean start in a laminar.

You are the kind of person that I need to visit and learn from - Or better still- have you here for a day to see what I have and how I do it- I have 5 HEPA filters 0.3 micron in the system, the building is positive pressured with a pressure bias to the lab and growroom meaning that the fruiting room cant backflow to the previous room. Its all controlled by a Cloudline inline fan and control system. Im rather proud of it!
This is a great response. I would also suggest you look at some of the low tech methods and buy spawn initially. This will allow you to focus on production sales and marketing. You can then backfill the spawn production. The building won't be helping. Working in IT in food factories the biggest problem is what the cleaning crews do overnight with the chemicals and high pressure hot water.
Im just gobsmacked that virtually all my agars get contam- then 75% of my grain spawn gets contam and finally whats left gets destroyed in the grow out bags. Ive been pouring money into it for 6 months and I hardly have enough to get by on now. my savings are nearly gone.@@stephengreaves7935

Im way past that Berserker- had three prescription antibiotics that were ineffective. Im now on MMS and DMSO mouthwash then coconut oil and turmeric twice a day. Its holding back the infection but gum recession has been massive. Im allover YT watching dental hygene videos. All three of us have differing dental issues that are about 12 grand each!
Yes , look into overseas Dental holidays, Thailand is a great place for that as well for half the price if not 1 3rd you would get high-quality help. NZ and Oz Dental prices are criminal. I think this needs to be addressed by the new Govt but that's another conversation. Best of luck.

I have no idea but my brain works in mysterious ways and sometimes solves problems that I have no basis for knowing. The only thing it's saying is maybe you're too clean?!? It doesn't make sense to me because I know nothing about mushroom growing (mycology?). I have heard of a couple mushroom grows that were in truck campers, but they were in semidesert areas. But definitely not sterile environments or airtight or temperature controlled. Again I know nothing about mushrooms but that I can't eat them. As much as I like them, they seem to contain large amounts of the nickel which I am allergic to. Hope this helps because I'm in the same financial state as you are and can't help in other ways! I understand- and I agree about my own issues with the gum recession- I had a systemic illness for a year. My lovely lady that lives here now is a talanted herbalist and is guiding my recovery. Im so lucky to have her here. But the holistic approach to mushroom cultivation simply does not work. There is no bio-synthesis with these organisms - They have to be eliminated for reliable cultivation. No body else seems to have this issue so bad!
Is that sterile hood set up the way it is when you're doing sterile work? Put a glass door or curtains in the front. Set it up so that only your hands are in the hood. I think it's just too open, as it is now. Many years ago, I did sterile work for mushrooms in a large cardboard box with a glass top and holes cut for my hands in the side. I sprayed the inside of the box with lysol before working. That was good enough. Any contamination I had was from later stages.
Can you see the white coreflute panels on top? they get assembled as a hood when I use it. My next experiment will be moving to liquid spawn to grain to eliminate atmospheric exposure
Grind bakers clove's and peparica and powder the exterior of the building. By any openings! Maybe cover the intake with cheese cloth. Hope ut helps you get rid of them bugs! 💕✌️✅

Can’t help with the mushrooms but I have seen carnivore channels where people have stated that their gum and dentition issues improved for some totally cured. I think swishing with coconut oil stops decay. Worth a try?
Im using MMS and DMSO as a mouthwash 2x day followed by coconut oil and turmeric. It keeps the infection at bay but the gum recession has been massive. now my teeth are loose and moving and more food gets stuck there making the whole processs harder to fix. I honestly spend about 2 hours per day on oral heath practices
Thank's Michael. Yes that's what Ive done all my life as an engineer/ fixit guy. My shoulders are ruined. All I can do now is type a bit and some light gardening. Im so close with this project now I have to keep on. When this is solved Im straight into 3 grand per week.
Why not brew your own ethanol, simple sugar and yeast mash. If its for cleaning, turbo yeasts only takes 2-3 days. A cheap air distiller with a fan controller hacked into it allows you to set the boiling point. Not great for drinking but 92% is easily achievable.. No spare $$$$ I'm afraid.
Ive built a still and in a video very soon you will see my sugar cane just starting to grow. I planted 20 growing shoots just yesterday. Here in NZ we will experience a sugar shortage soon because of shipping difficulties
@@t.h.o.r. Replace the sugar with a starch (potato) or fruit and ya good, a lot slower but doable. Have you considered Sugar beats, Most of our sugar in the UK comes from the beat. By product is cattle fodder but not the highest yield.
yup 30 years back I watched a gov sponsored research into beet as a sugar source- they sunk loads of money into feasibility studies branching into ethanol fuels- biomass gas generation and syrup production. From a south Island farmer and supplier on agar plates- his remain fine throughout their use time, mine blow up with contam 3 days after being made. Ive changed the way I make cook pour and store in so many incremental steps- yet still the mould!

For me it looks like different types of Yeast! candidas. Is the Agar steril? You could close the Plates with Parafilm. around. But I did not really get what you are doing.
I leave them in front of flow hood or in sterile still air box for a while to settle the condensation as much as possible. then I parafilm them for storage or growout. The Agar should be sterile- 200ml batches pressure cooked for 40 min- cooled and poured in a still air box or in front of flow hood Im getting various contam at every transfer! the building was waterblasted and disinfected then lined with insulation and plastic lining. the air system has a total of 5 HEPA filters the air exchange is 20x per hour
