You are welcome. The whole point of defining threshold is to select the specific area of interest for analyzing. The image may contain debris or noise that needs to be excluded during thresholding. Performing a correct thresholding is necessary to maintain accuracy of quantification. There is a debate amongst the scientific community, whether to keep the thresholding value same or different across all the images during analysis for comparison.
