Love seeing that trigonelline might be doing something. Based on your results on April 29th, I might try out some clover sprouts too. I'm also not particularly interested in adding in coffee into my diet.

Thanks @mervynman6303. I eat the majority of the strawberries frozen in a daily smoothie with ~330g raw beets and 60g fresh parsley, which are nutritional All-Stars.

if u ever do experimentation with coffee, then u can add in ur coconut butter (of course without altering total coconut butter intake) and blend it and make a pretty nice tasting bulletproof coffee equivalent 😋

Nice work as always! About trigonelline supplements: it seems that Nootropics Depot sells Fenugreek extract capsules standardized to 2.5% trigonelline, so that could be an option if you decide to supplement in the future

Interesting, and thanks @the123apz. 3% trigonelline = 30 mg/gram capsule. That would be 4 capsules for this recent test to yield 120 mg trigonelline, but 8 to equate the trigonelline content coming mostly from clover sprouts for the next. In other words, a lot of pills to match what I can get from diet...

Definitely James. Plasma levels have been increasing over the past 2 tests with the inclusion of trigonelline-containing foods, with minor effects on biomarkers, though (so far)

Great video! Always learn something new! Is the main reason you lowered nicotinic acid because of it's impact on LDL, or was there another reason? In other words, why move away from NA if it was substantially increasing NAD levels?

I always try to optimize biomarkers via diet as a primary strategy, so if I can do it without nicotinic acid, that's the goal Low-dose NA may aggravate my GI tract in a similar way to tryptophan-I'm correspondingly doing that experiment by taking it out here and there while testing NAD levels...

Mike, I love your channel, your research and the way you present . I bought red clover seed on Amazon and just grew my first batch of sprouts 4-5 days. Are they supposed to be yellow? My broccoli sprouts are green. Thx.

Hi Ed, and thanks. They might be yellow for a day or 2, but switch to green around day 3 or 4 btw, recent NAD results are in with 2x clover sprouts, and NAD definitely didn't increase. There may be a U-shaped curve for trigonelline, I'm not yet sure...

On coffee trig, not sure if the data is on bean content or beverage. Will have to run that down. My trig level is good per Iollo (1.5 micromolar) and I do drink ~ 3 cups per day mostly decaf. Plant based diet (kidney beans etc)

Hey Peter, my plasma trigonelline level increased to > 1uM (1.1, 1.7) for the last 2 tests (previous 5 test avg: 0.3uM) in conjunction with the small NAD bump for the latest test. I wonder if your NAD would also be ~30uM with a similar plasma trigonelline level, but other NAD precursors can impact NAD levels, too, so also maybe not.

This is a question that befuddles me as well, also in conjunction with the research on other coffee health benefits - in which case it mostly is about the beverage. However, I don't particularly like the beverage, but can eat some roasted beans - which raises the question about the correct dosage (I get jittery from a much smaller weight of beans vs weight of grounds in a cup) and whether the same health effects apply, given the differing preparation.

@@LVArturs I drink 1 cup caffeinated and two cups decaf. Don't think I'll risk beans straight. As for being jittery I think if you take L-theanine along with caffeine you might avoid.

@@conqueragingordietrying1797 Nichola Conlon has a couple good interviews on Siim Land's channel if you'd like to get a taste for her thinking about boosting NAD. The ingredients in her supplement are just posted out in the open on her website if you want to experiment with them individually or research them individually. (e.g. she likes nicotinamide over nicotinic acid).

Michael - good info. Crimson clover seeds you have as highest amount of trigonelline. Can use these? I see can get organic 5 lb bag online for only $24. Use grinder for seeds and put powder on salads, smoothies, and use as maybe spice over cooked veggies.

Do you know that red clover contains isoflavones, plant-based chemicals that produce estrogen-like effects in the body? As a peri-menopausal woman, I am very familiar with that supplement, and I know very well what effects it has, does it affect male body the same way? Not sure, probably not but I hope you did your research on that aspect of red clover supplementation.

Does niacinamide raise NAD to any notable boost or does a person have to use the flushing kind of niacin or NMN? I remember someone saying somewhere just taking some ribose with the niacinamide is good enough to get NR forming dunno if that is true.

Niacinamide does raise NAD, but it's been reported to increase homocysteine more than nicotinic acid, which is why I've focused on the latter, not former.

Great video. I did a Z test and the resulting p-value that your latest measure is just random variability is .0455, if we take all your previous no-treatment tests as representative. Scraping that 0.05, things are looking good. Where did you find crimson clover seeds tho? all i see in my country are seeds meant to be planted, are those edible?

red clover is Trifolium Incarnatum. ...and that is a unique name. It is pretty cheap (if available), here it costs 8€ per kilo, and sprouts are clean

It seems clear that you can manipulate the amount of NAD in your blood with various supplements or foods. But how do we know if it is actually increasing the NAD, not in the blood, but in the cells where are you to do you good?

There's published data that blood levels can increase in conjunction with muscle levels: pubmed.ncbi.nlm.nih.gov/32386566/ We'd need muscle biopsy to confirm, which is impractical (and I don't want to donate muscle to prove a point). To try to get around that, metabolomics (iollo's kit) can provide insight about what may be happening inside tissues, so I'm sending blood for NAD and metabolomic analysis on the same testing day

Before supplementing with any form of niacin or substance that may be converted to niacin at some point, you should read the study about what happens to excess niacin in the body. In short, it could significantly shorten your life. It appears that excess niacin is converted into the substance 4PY which is strongly associated with heart attack, stroke and other adverse cardiac events. A lot of people these days are doing unnatural things to their bodies without the data necessary to evaluate the likely long-term effects.

Why did you lower your doses of Nicotinic Acid? Appreciate what you're doing sharing this information. I would like to suggest making the presentation a little clearer, it's really messy to look at and figure out what's going on, like trying to decipher some rough annotations, I'd go as far saying it makes what you're saying more difficult to understand.

Nicotinic acid is a potential confounder when testing the clover sprouts-NAD relationship (if it exists). I fixed that for the next test, removing nicotinic acid. Thanks for the suggestion!

Okay, Mike, now you're making me nuts. I'm already growing parsley and dill greens in my Brooklyn apartment, and now I"ve gotta find red clover seeds. I'm not sure if this channel is moving the organic seed needle---but I just went to Ture Leaf Market, the company I buy my microgreen seeds, and all they have left is 1 Oz. packets of organic red clover seeds. Where are you finding red clover anyway? Aren't you here in New York? It's not like someone can just breeze into Whole Foods and pick up a pound of clover sprouts. I know you are as busy as hell, but maybe you should give microgreens a try. I've been very successful growing radish, broccoli, arugula, dill, and parsley with just microgreen trays and grow lights. For the coffee thing maybe you could supplement with green coffee bean extract. I'm taking that also. Consumer Lab has the Vitacost brand as a Best Buy, and they are pretty inexpensive. I guess I'll give growing red clover a try. Thanks for the video. I saw your last video with Chris. You look very young for your age. I guess it's working!

@@conqueragingordietrying1797 Well, which variety of clover from Ture Leaf are you buying? I'm on the site right now, and there are Mamouth Red and Medium Red Clover seeds available in pounds. But the Organic Red Clover is only available in small pouches. Hmmm, that's the same situation with Johnny's Seeds, too. See what you did?! You posted this video and sold out the entire organic red clover seed bank in North America! You're a Boston guy? No accent. You really seem like a NYC area guy to me. I liked the point you made in the last video with Chris about this VO2 Max protocol only giving us a chance to live to mid-80's, maybe. The Bible says Moses lived to 150! And I guarantee Moses wasn't pumping iron or training for any marathons. My grandmother sat by her 3rd story window in the Bronx smoking Marlboro Red after Marlboro Red to the tune of 4-packs a day and lived to be a healthy 94. But, she survived colon cancer in her late 20's and ate almost nothing every day. I guess if you wear a colostomy bag eating anything probably doesn't seem ideal. The only thing my grandmother ate was cold cuts from the deli on her block. And even with that she'd maybe eat a few slices of roast beef and swiss cheese. The only vegetable I ever saw here eat was a few spoonfuls of potato salad. I think that CR (Calorie Restriction) is a powerful tool. I've been watching Peter Attia for 10 years and He gets carried away with his exercise regimen when it comes to the average person. I'm a certified Health Coach, not charging anyone at this point, and getting people to just add steps to their day or take up walking is asking a lot. Thinking people are going to stat pumping iron and worrying about VO2 max def isn't for the general population. I also get pissed at his....what I think...is over the top protein recommendations. He Don Layman on---who has connections to the cattle industry---and Don Layman is like, "Eat two cows and call me in the morning" when it comes to daily protein intake. It's too much! Attia made that minny clip where he says "I can find no evidence that high protein intake correlates with a higher risk of cancer". That's probably not verbatim, but Jeez Peter I can easily find a direct correlation between high protein intake and cancer in reports written by NIH. Anyway....hope you find the clover seeds. Its super cool you are growing your own sprouts. If you haven't grown microgreens yet be careful its addictive. Plus, the leafy greens anyone on the East Coast buys from a grocery store are best case scenario already 4-5 days old. It's actually better buying frozen. You lose the fiber but you get more vitamins. Later...

@@conqueragingordietrying1797 Perhaps it gets metabolized or used up in some pathway while the seeds sprout. On the other hand, during coffee bean maturity, the contents of trigonelline changes very little with coffee seed development, so it might be the same with clover sprouts. BTW, Trigonelline is present in green coffee at 1% dry weight, so this could be another source. During the roasting process of coffee beans, trigonelline changes into N-methylpyridinium and nicotinic acid as its major products, which makes it a useful index of the degree of roasting. Normal samples of roasted coffee contain from 95 to 263 μg./g. of nicotinic acid. The nicotinic acid content of coffee is not dependent on the variety of bean. So you may ant to check its effects on your NAD levels.

There is published data showing that blood NAD levels are similarly increased relative to muscle when NAD levels are restored with niacin: pubmed.ncbi.nlm.nih.gov/32386566/ Outside of a muscle biopsy, intra-tissue levels are impossible, but to try to get closer to the truth, I'm sending blood for plasma metabolomic analysis on the same day as every NAD test, which can provide insight in terms of what be happening in tissues.
