Hey Friends,
the indirect ELISA works with two sets of antibodies. A primary one assures specific binding of the target antigen. A secondary antibody recognises the primary antibody. The secondary AB is linked to an enzyme converting substrate to a measurable product.

What does the strengh of the colour indicate?

According to video we checking antigen present in sample hence, the more intense the colour the no. of antigen present is more in the sample. ( as the more antigen = more antibodies binding = more binding of enzyme linked antibodies = increase in product(reason of colour) of enzyme and substrate reaction = more intense colour. )
