After rejecting the instant pot for sterilization method for years, I finally decided to give it a go. If you watched my last video ( • Grain Spawn in the Ins... ), you saw that it is definitely possible to successfully sterilize rye grain in an instant pot! The point of soaking the grain over night is to allow certain organisms to germinate, these organisms while in spore form can tolerate 15psi, at 90 min but once germinated they cannot.

Thank you for the piece of information, Iv just did my first grain pasteurizing and soaked it for 24 hrs following shroomy guidelines, didn’t know why till you mentioned it, will do that from here on out as it makes sense.

@@irvvvv601 u know that's total bullshut what he just said. People like making extra unnecessary steps and calling it endospores. Everything dies once pc reaches a given Celsius nothing can survive thats the whole point of pressure cooking. This what he's mentioning is a old man's tale. This is why we call it sterile.

I’ve had more problems with splitting grains causing contamination. I’ve been doing No soak no simmer for a while now and if I don’t get split grains I’m less likely to get contamination than from the 24hr soak prior to cooking

The link to the step by step didn't work for me. Here are my notes 1. Pour 1/3 Cup of Rye Grain into Mason Jar 1. OR (90 Grams of Rye grain for each pint jar) 2. Once you have the right measurement of rye for as many jars as you are going to sterilize. Remove grain from jars and place into Instantpot. 3. Add water to cover rye grain 4. Use “saute” option on instantpot to heat up grains until boil. Boil 1 minute 5. After boil, drain grains in colander, and let sit for 10-15 minutes 6. Pour grains onto sheet to and spread them evenly to allow them to dry. 1. Grain should not “look wet” should not be shiny. Looks dull, but is soft. (Took video about an hour and a half to dry) 7. Fill jars about half way full 8. Put your custom lids back onto the mason jars 9. Put tin foil ontop of mason jars to avoid extra moisture getting into the lids. 10. Put something on bottom of instantpot to avoid letting the glass jars touch the bottom of the instantpot, put few cups of water in instantpot 11. Place jars in instantpot, and select high pressure, for 1.5 hours. *disable keep warm button* 1. Guy in video did this right before bed so by the time he took the lid off everything was nice and cool. 2. Jars can be used within 1-2 days after sterilization for max results.

So i have an instant pot but it doesn't have the pressure cooker button or a yogurt either any suggestions? I have an instant pot IP-LUX the pre-setting options that it has is rice, multigrain, porridge, stem, slow cook, sauté, egg, cake, Meat/Stew, and Soup/Broth.

Can you standardize the moisture of the grain by weighing it after cooking it to get to a specific hydration rate? If too heavy, let it dry more? Curious what the hydration rate % should be if using this method.

I’ve seen some people do this with popcorn. They weigh 10 cornels dry and divide the weight by 10 to get an average weight. and then keep hydrating until they reach their desired hydration using the Same weighing method of 10 cornels and dividing not sure which is the right number because I haven’t experimented myself.

I'm new but I believe you can actually wait longer maybe up to 7 days but the longer you wait the more likely a contaminate will grow in that substrate. Even in sterile conditions there's a chance that 0.01% contaminate might survive the sterilization method or that something got pass the sterilization method but if it had been inoculated the mushroom will help keep the substrate free of other contaminate.

I’m about to do all of this Also. Can you tell me how much water to use on the step when you are pressure cooking the jars. I have the little thing that comes with it. But when I did this last time I ran into moisture getting into the jars(even wrapped them like you). But I think some of my jars seals may have been damaged when I was adding the injection port. Anyway I caught some tric in some (2) so far. But I am about to adopt this method lmk about that water and thank you for your work bro really!!!

Totally not a precise method thats not easily repeatable. Everytime you do this itll come out different. You need to figure out the exaxt amounts of grain,water, other additives so you can repeat this 100% the same everytime. Not put about 2inches of water in there. What if my pot is deeper?

As many people have said this definitely works. My spawn jars were in 30+ degrees Celsius where contam is even more likely for over 2 weeks. Works great

Thank you. I've been avoiding grain and just biting the bullet of pre-made grain bags, but it's been 50/50 on if they colonize at all... this is simple enough to do, and should make the failures a lot less costly at least

A couple additional notes on using an Instant Pot for sterilization...which I love. It is important to understand that altitude impacts the max pressure these devices can attain. Some manufactures compensate for this, but the info is rarely published. This can help account for the varied experiences people have with these pots. Instead of guessing, a $6 roll of autoclave tape will tell you if you are getting the temperatures you expect. While the tape is generally used to measure the outside of a container and cannot guarantee sterilization of media, it can be used to determine a) if you get to sterilization temp, and b) how quickly you get to that temp (using some test runs) This can inform how long you need to let grain bags or agar cook with your pot at your altitude. I'm at 6500 feet and began wondering if my 6 qt. Instant Pot Duo (that I use for everything mycology) was hitting the temps I expected. The tape confirmed that I could trust the pot. I've not tried to insert tape strips into the center of test bags yet, but a quick Copilot chat tells me this is a valid protocol.

Hey this is a great video people should realize that instant pot really is just a small pressure cooker and I really don't think you need to soak the grain overnight because that kind of defeats the whole purpose of the instant pot. of it if you're gonna cook Pinto beans and other things on the stove you soak them overnight but having an instant pot means that you can throw dry beans and water right in the pot and you're good to go this applies the same thing for the grains you're prepping for the jars.

its to deal with endospores. You need to presoak so they get killed in the pc process. don't skip the soaking.

Many people recommend venting the PC for at least ten minutes before putting the rocker on... Would it be necessary to leave the instant pot on vent for a similar time period and then seal it, to remove all the air inside and replace it with steam like in the PC?

is the lid with injection port on the cover necessary for sterilisation in the pot? or can i just use normal, completely sealing, covers? (because i have grown the mycel in agar petri dishes and plan to just open the lid and add the agar and mix.)

When you are done with the 90 minute sterilization in the instant pot, if you are trying to speed up the cool down time, is it ok to do the pressure release at 90 minutes, or just wait until it naturally releases as it cools down?

Definitely let it cool down slowly, every resource I've referenced says that rapid cooling increases contamination likely due to the fact that the thing cooling it (the air blowing over it, or the water) will not be sterilized itself. It is most sterile when not exposed to factors outside the pressure cooker.

@@andrewedwardsmusic if your contents are properly secured in jars/bags then blowing air wouldnt have any affect, however depressurisation can cause forced entry through gas exchange ports you might be using like micropore tape or micron strips

After pressure cooking and having the same jar setup as you, I always get a ton of moisture in the jar even if I completely wrap them 5 times in foil. Is that ok? After a couple of days, the grain seems to absorb some of that visible moisture in my jars.

I heard that any excess moisture that come from the pressure cooker is OK, you just shake it up and distribute the moisture throughout the other grains. I’m new to this, so I’m not sure if that’s 100% true but that’s something I have heard.

Whole oats expands 2-3x bro an instapot is a pressure cooker idk why hes acting like its not. I always do quart jars 1 at a time. Lay it on its side and do 2H30M with oats. I use oats and an instapot 0 contams

If you figured the dry weight to wet weight ratio, you could use the exact amount of water needed in the sterilization process. Basically, eleminating the1st half of your video.

I found I had a problem with my Instant Pot, after cooking one batch of grain jars and letting them cool awhile, when I added 4 more jars it would appear to boil but not get up to pressure~~~I found that I had to dump the leftover hot water from the first batch and re-fill it with coldwater to get it to get back up to pressure! Then everything worked fine. There must be some kind of sensor that needs to be reset with cool water before it will allow the water to heat up to pressure temps. Has anybody else had this problem?

I can’t fit my jars with the trivet, quart sized, so I don’t use it. No problem with breaking yet and I do 3hrs typically for my rye jars.

You actually could inoculate through the patch, just put a new piece of tape over the hole after inoculation. I install a rubber plug in my lids to allow for inoculation through the plug without disturbing the vent.

In theory yes. Personally I never had problems like this. Might also be because instant pot takes a long time to build up pressure already. I think it also matters how big your substrate is. If you have one solid grow bag inside, it will probably take longer for the heat to reach the core.

So that extra 3 psi you get from standard pressure cookers isn’t necessary for mycology sterilization? Do you know if this method is enough for making agar?

liquid mediums are much easier to sterilise, so if hes making grains work with the instant pot then agar will work as well. sorry i skipped through this vid mostly but also 15psi for 30m is enough for agar. there are nice graphs on google images to show sterilisation time to PSI. 12psi requires 3h and 15psi around 80-90m. The is it necessary question really means can i get away without contam, sometimes yet but sometimes no

I'm not sure what a Bailey jar is! I think your best bet is to look at the dimensions of an instant pot (available on their website I believe) and compare that against your jars.

No. An instapot or pressure cooker builds pressure and much higher temperature. A crock pot will never get hot enough to sterilize anything.

Robert, I don't want to give you an answer without giving it a try. I suspect the answer is yes, but I'd like to do some experimenting. The IP is pretty small though, so I don't see it being possible to do more than one 5 lb bag at a time.

You need a special heat resistant plastic, I've seen it called PP5, and I think it's recycling designation is also "5". Ordinary plastic, like g Glad sandwich bags or freezer bags will melt.

There's controversy about Unicorn plastics no really being biodegradable, but just disintegrating into microplastics. The EU won't certify them, but they claim this is a conspiracy by European industries involved in more usual biodegradable plastic tech. IDK

I put the lids on until the ring stops spinning without me adding any additional effort. I don't put it on tight, or loose....lol hope that makes sense. And I agree with @Andy Sills, it doesn't matter much.

​@@OneEarthMushrooms ​ Thanks for the reply, Jared. I found the study over at the @Mycology Exploration channel as they also advocate an Instant Pot. The study is titled: Assessment and verification of commercially available pressure cookers for laboratory sterilization and was done @ Dakota State in 2018 A Bing search will bring it up. One of the pots, the Duo 80 I believe met their standards for lab sterilization. Thanks again for you interest here...It is great to see a knowledgeable grower exploring some of the topics that were discredited in the past. As materials and equipment change, we need to update our knowledge and teks accordingly.

There's a lot of sterilization advice that's way off. I believe the standard for medical autoclave sterilization is quick, like 20 minutes to a half an hour. Of course InstaPot is a big step down, but... if you run pressure cookers for 3-4 hours, of course your not going to have contamination problems! The point is to do this as quickly and efficiently as possible.

You're looking to buy the grain? I don't have any links. Depending on where you are in the world you may be able to find rye grain (not rye seed!) at your local farm store. I personally buy in bulk from Azure Standard. Takes a while to get the grain shipped, but its really inexpensive for high quality grain.

Search for 3M Micropore Tape. I've bought it on amazon before, but a local medical supply store would be your best bet.

Question for the educational and theoretical purposes only Lets say you have an "All-in-one" grow bag and several 1/2 pint jars that are almost 80-90% fully colonized, but you have just been invited to go to, let us say Mexico, for 10-14 days(starting in 5 days..so around >>14 days past being fully colonized

How was Mexico? In the bag (or jar) it will be okay, even if it eats all the grain in the container. Once you take them out to fruit, presumably in a fruiting chamber, you can't leave it, at least not without harming it.

Hey, great video! Thanks for high quality education on mushroom cultivation. Question - why do you use 4 jars? Isn't it more convenient to inoculate just one big jar? Is this because 1 won't fit into the instant pot? If you're growing the same mushroom, do you use multiple jars to have enough spawn for the 1:4 spawn -- to -- substrate ratio, if using a monotub? Thank you!

little jars fully colonize a lot faster. and if there is a cotangent in the one Big jar you must start over. If one of the little jars is contaminated you still have 3 others.

I have been looking Everywhere for a jar larger than a Pint that has a screw on sealable lid of some sort, practically impossible. If you want it to fit specifically in an instant pot. Pint size it is.

This is true for most stovetop pressure cookers as well, although you can try putting a big jar sideways. Maybe canning pots or high end pressure cookers can take larger.

I came across an even simpler method on Shroomery. It comes down to using the exact amount of water for the rye. Put 2/3 cup of rye berries and 3/4 cup of water directly in the jar, seal it and sterilize it in Instant Pot (I use the IP's Sterilize program for 35 minutes). Done! I've been using this process successfully for a few months now.

@@HelpMe_OrNot For grain jars I make lids with only the injection port and a simple GE filter disks. For Liquid Culture lids, I use the bigger plastic "injection ports" with built-in GE filters. I don't use it for injection (the mycelium wouldn't make it through the filter) but they are more resilient to contam by liquid than the little filter discs. The grain jars colonize within 5-7 days for most Oysters but a jar of Pink Oyster last week took only 4 days to colonize!!
