don't you want to pour media into the dish first and let it settle before adding your inoculant? If you do it in reverse order like you do in the video you maybe burying your inoculant under the agar media and deprive it from oxygen.
@@SoudaAlvi I would say pour agar first, cover with lid, let it cool. After it cools - add your culture onto surface of agar (make sure you wear a mask, gloves, etc. perhaps use a flow hood if you have access to that). Then close the lid and flip it upside down (agar on top, lid on the bottom).
@@SuzannesTutoring Also agar could be hot enough to kill some cultures, as you know pasteurization sometimes requires fairly low temperatures like 60-70C.
@@tolibelom sir I will do any isolation first time (in my life ) of bacillus licheniformis from soil sample. I have some confusion. Please rep I will do serial dilution . 10ml distilled water is present in 1st tube. I want to ask ,, directly add 1g soil sample in first tube after weighing (mean dilution make in ist tube itself when we add soil (solid) of 1g in first tube) Sir my 2nd ques is I don’t find selective media for bacillus licheniformis I grow it on LB agar in coming Monday. After serial dilution I pour it on agar and incubate it for 24 hrs. Then what will I do¿ it’s biochemical test. Or I transfer it on another culture media of same type which is LB. ( after 24 hr, I see any colony which show different shape , colour which is on same plate before , I transfer it on different plates by streaking on new agar. Mean all colonies I see transfer it separately on different plates. My 3rd ques is I want to do heat shock treatment so that only spore formers will remain in my sample. I will keep my sample in hot air oven or water bath it for 70 Celsius for 10min. I want to ask directly do this procedure after sample collection . Only keep soil sample in water bath or make dilution¿ My last ques is sir my bacteria is anaerobic. So i do serial dilution like this sir did. ( my bacteria will not die due to this…¿ Or after that place petri plates in anaerobic jar. Mean what preventive measure I follow for doing serial dilution.
Observe under microscope. These will be of different shapes. If they're curved shape then they will be vibrio cholerea that cause cholera disease. If they are in helical shape then they will be spirillum volutan. If they are in chain then they will be streptococci. If they are in pair shape then they will be diplococci. If they are in spherical shape then they are cocci and if they are in rod shape then they are bacilli.
