I always wanted to know how Dio would have fared against the pillar men, since vampires were like food ti the pillar men, but Dio was not the average vampire

What do you mean? Dio was always an average vampire, he had nothing special, he just looked stronger because we saw him in Jonathan's perspective, as you said, Dio is just food to the pillarmen, if you touch them, you'll be absorbed

I haven't played this game, but how do the other characters walk across the spikes in that map? Dio and the Pillar Men make sense, and so do Jonathan and Joseph because of Hamon, but what about the others who aren't "immortal" or use hamon? Does the game basically just pretend the spikes aren't there? or...

From what I remember, they take constant damage when they walk on the spikes.

Vampires and the Pillar men are completely immune to the spikes, Hamon users are immune if they have the hamon bar charged up, though it will deplete it. Everyone else eats shit
