Security default cannot be enabled if you have conditional access. In that case ‘manage security defaults’ is replaced with next text ‘manage conditional access’ what redirects you to the page of conditional access and no longer to page security defaults
You can't disable MFA if you have Security Defaults enabled, so the break glass account wouldn't work as you explained. Maybe it's different with an E5 License but I don't think so.
You can not use a break-glass-account if enabling security defaults - that enforces MFA without exception so it doesn't matter if you have 200+characters in a safe if you don't have other "factor" available when you accidentally broke anything.
