For Morooo, being a student of genetics.. the concept is pretty basic... That DNA present in all the cells are actually same and identical... But difference occurs because of expression of genes on DNA (as all DNA is not useful only certain portion act as a gene). For example, finger cells have more touching sense genes expression, nose cells have more smelling sense genes expression etc etc.. but overall DNA of all cells remains same. And also variation doesn't occur in normal division of cell like in skin or any part.. because it is mitosis ( cell division without variation) so they are identical copies.. and variation only occurs in germ cells i.e sperms in case of human (meiosis, cell division with variation). And as far as CRISPER is concern...it is gene editing and words itself is saying it is editing... So modification or variation occurs but at certain selected and specific point, not all DNA.

Genetic disease are due to some amino acid in a peptide chain which is abnormal for example in sickle cell disease there is there is valine instead of glutamic acid… This is due to some very minute abnormality in genetic code which translates in to abnormal protein which fail to function

@Mooroo bhai you know your genetics don't change with ageing there are two type of genetic sequences coding and non coding, so the current most acceptable theory for aging is that of telomere shorting. These are repeats of non coding sequences at the end of your chromosomes, they degenerate with each successive division as they dont code for any protein so even if they do degenerate your genome remains the same still. Ageing is still not fully understood but its on of theory in genetics.

Your DNA sequence remains the same - Muhammad Khan is correct about that. You stem cells (mother of all cells) and terminally differentiated cells (a final stage) skin, lung, blood, brain, etc. have a same copy of DNA, difference is terminally differentiated cells express different genes at different stages of life cycle via epigenetics and external or internal stimuli. This gives them unique function. Stem cells on other hand constitutively regenerated themselves and differentiates into terminal cells throughout your life. Genetics is amazing, and we're just scratching the surface.
