فيديو مفيد جزاك الله خيرا ممكن الخطوة التاليه. مثلا انا عندي 20مركب مصنعه جديده اوحتى مركب واحد،أريد افعل لها سكرينينج, كم الكمية عشان اخذ من هذه المركبات ، وكم ميديا اخفف بيها مثلا رح ابد بتركيز um 20أو 50um البعض يستخدم هذا المعادلة c1v1=c2v2 كيف احصل على المعطيات .وكم الكميه من المركب والميديا عشان أضيفها على ،96plate Mtt experiment

شكرا لتعليق حضرتك حاضر هسجل فيديو جديد لاضافه المركبات للخلايا عامه التركيز معاك 10mM 10000 uM x V1 = 50 uM × 200 ul media in the well V1=(50x200)/10000= 1 ul of tested compounds (0.5% DMSO) Or V1=(25x200)/10000= 0.5 ul of tested compounds (0.25% DMSO) 0.5 ul is the lowest volume that you can use with 0.5-10 ul micropipette unless you have 0.1-2.5 ul micropipette Best of luck If you need further assistance, contact me via WhatsApp +201551919531

Thanks Dr. for your valuable video, but I want to ask what if the compound is water or Saline soluble, is there a recommended concentration to be prepared and stored in -20c like DMSO 10 millimolar? or I prepare any stock bigger than the concentrations I need to be able to dilute. and what about the control one for cells, I use the percentage of water or Saline dissolved the compound to media like DMSO? thanks.

Dear Emi, Thank you for your comment. You can prepare the highest concentration that you can achieve from your compound. It depends on the solubility of your test drug. You may solubilize it in water or the culture media without serum. The stability of your compound has to be evaluated in water. If it is stable, store it at -20 or -80. To be in safe side, aliquot your solubilized drug to avoid thaw and freeze cycles. If your test drug is soluble in water, this doesn't allow for you to add big volume to the cells. The permissive volume is between 0.5 to 2 ul ( you have to be careful not to change the buffering of the culture media and it's components) For example: My compound is soluble in water I will add 10 ul of my drug to 190 ul of media In this case, there is a change in the components (20%) of the media that will affect on your cells. If you will visit the NCI website, you will find that they use just 0.5 ul either from DMSO or water soluble compounds or natural products. dtp.cancer.gov/discovery_development/nci-60/handling.htm Best wishes

thank you for your helpful video, I have a question cisplatin solubility was reported as 1mg/ml but the best solubility I got was 1.5mg/ml with concentration 5mM It was my stock I need concentration 100 UM 5000*? =100*200 4 micro liter from the stock Represent 2% from the solution. u kindly said I must not exceed 0.5-2ul from saline to media (0.5 - 1%) Is it acceptable to use saline with this ratio or how can I modify this value

@@amiralsarhan7694 I prepared cisplatin in saline solution as (8 mM). The highest concentration was 50 uM in the well

شكرا كثير لحضرتك دكتورة ربنا يجزيك كل خير.عاوزين مثل ده الشرح الرائع لو الطالب حضر مثلا سبعة مركبات عضوية في المعمل وعاوز يدرس فعالية المركبات ضد مثلا اربع انواع من البكتيريا وضد مثلا ثلاث انواع من الفطريات وضد مثلا الاكسده .عاوزين نعرف شرح التكنيكات والطريقة مثل الفيديوهات الخمسة العملية اللي شرحتيها .والف شكر مرة اخرى
