You can prepare the highest concentration that you can achieve from your compound.
It depends on the solubility of your test drug.
You may solubilize it in water or the culture media without serum. The stability of your compound has to be evaluated in water. If it is stable, store it at -20 or -80. To be in safe side, aliquot your solubilized drug to avoid thaw and freeze cycles.
If your test drug is soluble in water, this doesn't allow for you to add big volume to the cells. The permissive volume is between 0.5 to 2 ul (you have to be careful not to change the buffering of the culture media and it's components)
For example:
My compound is soluble in water
I will add 10 ul of my drug to 190 ul of media
In this case, there is a change in the components (20%) of the media that will affect on your cells.
If you visit the NCI website, you will find that they use just 0.5 ul either from DMSO or water-soluble compounds or natural products.
