it can be incubated in normal tropical room temperature (25-28 degree celsius), in dark condition. Can you clarify what do you mean by filtre 0,2 micro?
@@user-fr7pv8qr6u yes, it's easier if the bag got a filter patch, but it's more expensive. you also can use a heat-resistant glass jar, make a hole on the cap, and then plug-in cotton wool... or use glass bottle, and cotton plug on the bottle's mouth.

Greetings dear friends I've made the entire procedure but I always get green mold. I used wheat grains instead of rice husk and glass jar, and the inoculation was made inside a laminar flow box. Can you help me to find the possible spots of contamination? bye
@@ropenedu Hi there🖐 1- I rinsed the grains in water, 2- I let it dry for 8 hours 3 -boiled the grains for 15 min, 4- Put the grains in glass jars 5 - I put the jars inside the pressure cooker for 50 min. 6- I let them cool one night. 7- I inoculated the grains with the petri dish culture inside the laminar flow box, then I put lids on with a hole and also put cotton inside de jar as a filter. 8 - finally i put the jars inside a big cartoon box, but 2 days later i have green mold around each piece of jelly with white mycelium that I put inside the jars Sir, do you have an email address in orden to send you some pictures? thank you very much for your attention🙏
Dear Sarah, it's a polypropylene (PP) bags, which can withstand high temperature with a melting point of 171 °C. Alternatively, polyethylene (HDPE) also can be used with a melting point of 130 °C.
It's ok for the production of spawn. However, not recommended to use food crops as the substrate. In addition, rice will retain too much moisture, which makes them not suitable.
yes. gypsum, calcium carbonate, calcium sulphate, etc. is used to increase the pH and act as anti-bulking. However, you can omit the use of these materials as well.

I usually use a metal tea spoon, wipe with 70% alcohol, then inoculate 2-3 spoons of the spawn. Otherwise, you also may use 1-5% based on the weight of the substrate.
@@ernestovillenajr9869 make sure the moisture content of the growing medium is good (not too wet or not too dry). Inculabe at 25-30 degrees celsius in dark condision.
I used an autoclave. Steam sterilization at 121 degree celsius for at least 15 minutes. but the whole process from heating, sterilization, and cooling will take approximately 2 hours. Alternatively, you can pasteurize at 80 degree celsius for at least 8 hours.
