I was diagnosed with mild scarring of my lungs in April. I came across a RU-vid vid of an experiment with mice using NMN, Sirtuins(Resveratrol) and Zinc. I started to take these NMN 1000mg, Resveratrol with Quercetin 1500mg and 30mg zinc every morning on an empty stomach. And with prayers. Yesterday, i had another CT scan on my lungs. The scarring has been resolved and disappeared! Praise the Lord!

I am almost 68 years old, and a lifetime everyday exerciser. I have also been taking 500 mg of niacin daily since the 1990’s. Nevertheless, I still developed edema in my lower extremities over the past few years. In addition, I have been subject to frequent calf muscle pulls since I started playing pickle ball daily about 15 months ago. Then, on October 1, 2022, I started taking one gram each of NMN and resveratrol on a daily basis. What motivated me to take these supplements was a particular Sinclair video where he spoke about the proven(in mice) ability of these supplements to promote growth of new capillaries (angiogenesis). Well, within two weeks of taking these supplements, my edema and calf muscle pulls stopped completely. That was huge for me. There is no question that these improvements occurred only because of these two supplements in the proper amounts.
NMN worked for me. After getting spondylolisthesis, from running marathons, I could no longer run due to pain (late 40's, in fact I couldn't even walk without pain). It totally changed my life (didn't change anything else around diet, etc)... Arthritis pain in my back is gone, better mental clarity, better energy, and no longer need epidurals every six months to function. Moreover, I can run again pain free and at the same pace 15 years ago at the same heart rate. I recommended to friends and family members and they've had similar results.
Can you still do that without ingesting NMN? If no then for how long can you stay healthy without ingesting NMN? It may be that anti-aging suppliments if actually effective will become just another everyday need just like drinking water or eating food.
I can personally highly recommend NMN from Renue by Science. I’m 53, male, physically fit, and a cancer survivor (prostate surgery and radiation). Last year I felt like I was 72 years old, from the radiation treatments. 8 months on NMN, and other supplements, and my stamina is all back to normal.
I’ve been taking NMN for about a year. I definitely have noticed more energy. I am a 50 year old female. I am a very slow occasional jogger but I was talked into a half marathon. the study you mentioned about NR was of middle-aged, obese, insulin-resistant men. As you mentioned in the beginning of the video, lack of exercise might decrease NAD. And I believe obese middle-aged men constantly have their NAD levels dropping. So the fact that by the end of the study their NAD levels remained the same might actually be a good thing, the NR coud've offset the NAD levels they are losing by virtue of being obese. For animal studies, NR and NMN have both shown to increase NAD in muscles. In a study on mice, NR supplementation increased NAD+ levels in skeletal muscle. Another study on mice found that NR supplementation increased NAD+ levels in the quadriceps muscle. In a study on mice, NMN supplementation increased NAD+ levels in the blood and skeletal muscle. Another study on rats found that oral administration of NMN increased NAD+ levels in the blood and gastrocnemius muscle. Proper human studies are needed, ideally not obese humans.
I wonder how much of his decisions are on faulty studies or metanalyses without looking at the underlying studies for flaws...too much info out there. Hard to sort through even just one field.
I am on 2 months on NMN and Resavertrol and i must say it has helped me from devastated chronic low back pain above 10 on the pain scale! I am alot better now in running which i could not run anymore. I was bed ridden! I have nothing to lose, I will continue to take them.

Backs as we age don't bounce back like we want to but if your able to walk keep moving. I have injured by back and thought I was OK and I pushed it to far. I’m a nmn user. My life has improved since taking it. I also started fasting til 6pm when I started taking it. I believe in placebo over physical medicine anyway sooo yeah nmn is fantastic.
I’ve been taking 125mg capsules of nmn I got from nootropics depot almost everyday for the past 2 months. Skipping days to see if I felt different(I feel much different now than I did before and I also stopped going to the gym but my muscles haven’t reduced in size much compared to my gym break muscle loss test I did during covid) I’m much healthier now physically and mentally. Over all I can say my constant thoughts of a meaningless existence stopped literally when I started taking nmn. But many things changed at the time so who’s to say. I do now live as if I could live forever so please try out that way of thinking and take nmn. Words words words

Sinclair does not profit from nmn but it seems his company are trying to capitalise on it through their development of their own version, MIB626, and in fact it was his company that caused the recent FDA ban in the US. As someone who has nothing to gain by telling people how good nmn is, I can assure you that it has saved and transformed my life.

If I stop taking it symptoms return within 2 days so I doubt it is placebo and I was really skeptical about initally taking it especially given the cost of the product. My dad started it a while back and got some energy back in the mornings, started improving in competitive sports that he was going downhill in and even quit coffe, he didn't change anything else in his lifestyle. May be a coincidence, but I think worth a try if getting results.

I’ve been taking 500mg Lipo NMN and 125mg Lipo trans-Resveratrol every morning (on an empty stomach) for about 6 days. I also take one NAD booster in the afternoon around 2pm which contains 125mg NMN, about 80mg NAD, and about 80mg NR. I must say my energy levels and mental clarity are greatly improved! I’m also amazed at how much better my mood has been. I’m not as irritable or depressed (I have chronic depression and GAD). Will definitely continue to take it!
I have Muscular Dystrophy. No one has any answers. So, I took it upon myself. I started nMn in February. I've gone from wheelchair to walker, to walking on my own. I'm not a scientist, but I have tried every supplement available. NOTHING has shown the benefit that nMn has.
@Streamlined955 I actually do. I was in a wheelchair 17 yrs ago. I read Dr Terry Wahls protocol. This Dr has severe MS. She began experimenting with diet and supplements. Slowly she began to move her limbs. One day she noticed she was able to stand and take a few steps. A few weeks went by and she went back to the chair and couldn't move her limbs. She looked at which supplements she missed if any. That week she noticed when she was off she hadn't taken the COENZYME Q 10. She was focused on the bigger supplements and was shocked when she put it back in she was able to move. I was diagnosed with CRPS ,CFS ,fibromyalgia and RH. I started NMN , I could barely get out of bed. The first 4 weeks I felt no difference. I began taking 250 mg of NMN with TMG,zinc and sodium bicarbonate. I went to 500 mg and eventually 1000 mg or 1 gram. It was the 4th month of taking 1000 mg ,Renue By Science that I noticed I was hopping outta bed,I was full of energy. I was sleeping well. There was a huge difference in my energy levels 1 year before and 1 year after. TBH I thought prove me wrong because it is expensive. I have more energy than I did 30 yrs ago. She is out of her wheelchair, she did trial and error with diet - it's a paleo diet best for MS ,and she'd take and remove the supplements. When she forgot the coenzyme Q 10 she noticed a huge difference, not thinking this little supplement could make that much of a difference, it did. I ordered Renue By Science with TMG NMN zinc and sodium bicarbonate. I started at 250 for weeks and nothing. I almost stopped because it's expensive, but I did go up to 500 mg and TBH I didn't see a difference. I was still exhausted, I have CFS .So it doesn't matter how deep or how much sleep I've had I constantly feel like I have mononucleosis. I went to 1000 mg with TMG, zinc NMN and sodium bicarbonate. It was the 4th month of taking it and BAM I felt like I had a ton of energy. I was shocked because I haven't felt this good in years. I am in a bit of a pickle now because I was taking 4 thinking the 1000 mg was all in one capsule as it has TMG NMN, sodium bicarbonate and the zinc.The ZINC was 20 mg in each capsule. Way to much. I am 63 and have had a number of heart attacks due to a fairly large hole in my heart which I have had since birth. NMN helps me feel better. However, after watching this presentation, I am considering trying niacinamide instead. If I get the same results I will stick to it.
Taking it for 2 years. When i stoped...i felt older...how can i explain...i was not the same at the gym....took it again and i feel like a beast. I take the lipo one from renew by sciense. So...i will keep taking it.
I started taking verso NMN & Resveratrol and I had a micronutrient test done before and after and there’s a significant improvement in my results. In terms of overall well-being, I made multiple changes in my journey so I can’t just attribute my improvement to this supplement.
What kind of resveratrol you take? What is it made of? Only type that seems to be sold here (in Finland) is made of Japanese knotweed and I'm not sure if it's any good.
I'm a 74 yr old male athletic runner/cyclist/resistance exerciser. I began taking NMN and resveratrol a year ago. I did not feel a significant difference in my running performance in the first few months. But my running began to feel harder about the same time. I began feeling muscular skeletal pain of sorts. I could not firmly ascertain exactly what I was feeling but I definitely did not feel strong and vigorous during my runs. This condition was consistent. My times dropped off significantly. I wondered if it was just an age related condition. But, I wondered why that would be the reason as I am a WFPB vegan with an astute sense of proper nutrition. I did not feel the cause of my decline in running performance was age. I discovered this channel just the other day, July 30th. I decided to stop taking NMN and resveratrol, one gram each on that day. My 6 mile run on August 1st was significantly improved equal to my running performance prior to taking these supplements. I realize it is too early to know for sure if the supplements were the cause of diminished performance. I have an intuitive sense that the supplements are the cause. I will update periodically. I want to thank Dr. Stanfield for his diligence and activism in alerting people to the facts.
People have unique chemistry and it’s very important to monitor changes in supplements and not assume it’s all good. I have gotten great results from NMN 500-1000 milligrams a day. Sinclair and others recommend taking TMG with NMN and I am not sure but that could be the reason for your issue. Either way it’s good you recognized an issue but your body needs extra TMG when taking MNM and it’s not mentioned in every video. For me it’s been a near miracle like others have said. It certainly will not “cure” everyone though.
To me, this isn't surprising. We are individuals with varying needs. The medical world keeps trying to apply a one-size-fits-all solution to all if us and it creates misery for many. Example: many are helped by various drugs for Afib. Those same drugs make my heart function much worse.
I have used both NMN and NR. I can't tell if NMN does anything for me. NR definitely improves my mental clarity a good amount, I'm impressed with the effect it gives me.
The benefit of the NMN/NR/NAD+ supplement I take, over the NR supplement I’ve compared it to, is that I feel more energetic and have massively greater exercise endurance. For example, I’m ordinarily too tired to exercise at all, yet on the combined supplement (which I also take with re rest of Sinclair’s stack) I was able to stand and shovel heavy gravel of 8 hours straight the other day, with only a couple of brief breaks to eat and hydrate, and even afterwards, didn’t feel the need to sit down. And I see massive improvements in HRV. I’m similarly testing Niacin, which also shows improved HRV, but makes my skin burn and itch to the point of distraction, so I can only take it at night, missing the exercise benefit. It’s the microbiome that is the missing conversion pathway by the way. I take akkermansia, and eat a whole food vegan diet, which is critical in the conversion pathway. The microbes that convert NAD+ require exercise and fiber to propagate.
I've spent a lot of money on supplements like NMN over the past year, which unfortunately I probably should've spent on gym memberships and massages. I have a feeling the longevity supplements field will be the wild west for quite some time. Thanks for helping bring us back down to Earth.
The large body of scientific evidence weighs heavily in favor of consistent moderate exercise and better sleep and healthy relationships being by far the biggest factors positively impacting longevity and health span. So you have the right idea. Despite all the hype and anecdotes, the evidence for NAD precursor supplements doing any long term clinically significant good is just about nonexistent. There isn’t a magic pill. Diet is a factor but it’s complicated. As a scientist, I don’t agree with everything Brad says but he is on target when he is saying that supplements are a secondary thing to lifestyle. They are a distant second.

I take it because I can't take Niacin and when looking at the options it was cheaper than NR. There is a battle over which is better NR or NMN .. both are NAD boosters and I think people should take whatever NAD booster that work best for them including Niacin. Taking mountains of these vitamins probably isn't really helpful its just a waste. It has most definately helped with the things that B3 is known for though.
@@sncnutrition7118 Niagen is the brand name for Nicotinamide Riboside Chloride. There is also a hydrogen malate form. Both of which can be found on reputable supplement sites like iHerb for as cheap as possible. You can price compare. If one has trouble with niacin, NR is probably the lesser cost no-flush B3. Though niacinamide as a no-flush would be cheaper. Just keep in mind, there are still a lot of unanswered questions and legitimate expert skepticism that any of this mega dosing of B3 precursors is really beneficial in the long term. At best it could all amount to being just a small 3rd order effect in terms of longevity or just useful in certain acute disease treatments. Optimistically, there might be a better answer in 5-10 years. Optimistically.
Update for NMN users - As we mentioned earlier, we had been experiencing embarrassing gas from our Toniiq NMN & Resveratrol powder. We attempted various approaches such as adding yogurt or not, blending it into a smoothie, dissolve in olive oil, or taking it directly under the tongue, but the issue persisted until we reduced the amount significantly. We suspect that the problem arises from one component being oil-soluble while the other is water-soluble, yet they are combined in Toniiq's formulation. It seems to be a clever solution, but it may not work for everyone. To resolve the issue, we have been experimenting with Niacin (FLUSH) and Resveratrol individually. We open the capsule and place it under the tongue, resulting in no more gas and being much less expensive. We hope this information is helpful.

It is generally not recommended to cut up pills on your own, but since Niacin is a Vitamin B, some may wonder how much harm it can do. Personally, I open my 500mg capsule and take 100mg at a time to avoid the flush shock, which can cause a temporary redness and itching resembling a lobster or a monkey scratching for about half an hour, in a light-hearted way. I used to put resveratrol under my tongue when I was taking Toniiq 3-in-1 powder, as it seemed to produce gas. However, I have since switched to Renue by Science Powder for NMN, which I believe is the main ingredient. Resveratrol is known for its strong antioxidant properties. To save money, I switched to lower-grade (50%) less filtered pills that appear brown instead of white. I swallow the Resveratrol pill and place the NMN powder under my tongue in the morning. I take the 500mg Niacin pill after food to reduce the intensity of the flush, although the flush is said to have beneficial effects. Sometimes, when I'm relaxing in front of the TV, I place a small amount of niacin powder obtained from opening a capsule under my tongue. Placing substances under the tongue allows for direct absorption into the bloodstream, bypassing the stomach where they would mix with other contents during digestion. This method is also considered to be easier on the liver. I would recommend looking into TMG (trimethylglycine) as a way to protect your liver. On a different note, the Niacin flush has been known to have positive effects in the bedroom department, if you catch my drift. I hope this information is helpful to you.

Thanks Dr. Brad, especially for discussing the exercise study. There is also work from Prof. Janet Lord which shows major immune system benefits of exercise in older people (including thymus preservation, which might be a partial consequence of staying relatively lean). Also work from Adam Antebi's lab showing that low insulin (in mice I think) raises plasma AKG and NMN, a lot. Other studies showing major benefits to overall health from simply walking more. I stopped NR (Basis) a couple of years and just make sure I get a reasonable amount of nicotinamide. Also make sure to get plenty of quercetin, apigenin, and EGCG in my diet as they appear to help modulate the NAD+ salvage pathway to avoid depletion.
That is a very good strategy - take much lower cost B3 and then other molecules to make sure the Salvage pathway is operating very efficiently. You should check out Nuchido Time. That is their strategy. They sell a supplement that includes Vit B3 and molecules that make the Salvage Pathway run most efficiently. Their website has a lot of good info and you can find a lot of YT videos / articles of the Nuchido CEO discussing their approach.

I'm over 60 and have been taking niacin for many years. I take one 250mg tablet, break it in half and take one half in morning and other before bed. Very mild to no flushing at all if you take it every day.
I tried NMN with some skepticism. I felt great, stopped for a while, felt much worse. Started again, feel great again. Not 100% sure it's the NMN of course but it does seem to work for me so far. Not sure what the mechanism is then. Especially since I'm young. My senses just seem heightened or something? I feel like my eyesight is sharper or something, smell is more sensitive. Brad have you ever taken NMN, just to see if it has some effect on you?
I think 100mg of Niacin is right for me as it gives me a slight flush but nothing too heavy. As far as the daily recommended amount, that is what you need just so you don't regress but not a value that will even give you the slightest flush. You are still very young though so you have nothing to worry about. I am 56 and I feel like I am in my 40s. I started counting my pushups in a row max a few weeks ago and my max was 50. Today I am up to 65. My goal is 83 as that was my limit when I graduated from Army boot camp and that was the 2nd or 3rd highest in my entire company.

What a fortuitous stumble across video. I recently ordered berberine from DoNotAge based on purity tests by another longetitivy channel. Only five grands out of 27 tested at 100% berberine. Hi Dr Stanfield, I hope you don't mind a question today: does it appear safe to take a combination of NR, NMN and Niacin together? I am 48 years old with high cholesterol. I currently take 100mg Niacin (any more than that triggers migraine), with NR and NMN. This is taken with TMG. I have found conflicting ideas about combining - some say that different forms of B3 may be helpful as different tissues might 'prefer' NR or NMN pathways - others say that combining is a waste of time. I have been taking it for about 8 months along with Multivitamins. Started getting bad rash on feet and hands, similar to what happens with overdose of Vitamin B. Terrible brain fog too. I wonder if your findings are right, and it is broken down prior to reaching blood stream, likely into Niacin. Stopped taking it and the rash is now receding. Slowly. The rash got really bad in the beginning, it hurt so very much. Thanks David Sinclair... Happy to discuss this with further. I’d be interested in finding the truth of this… Update - Turns out that the NMN gave me Gout, and the rash on the hands and feet is a rare presentation of the Gout (Uric Acid crystals). These can be formed by high doses of Niacin too. This makes it super clear that the NMN is broken down into Niacin in the body, and is overdosing the body in Niacin, which can cause Gout. The compounding issue was that I was not aware it was Gout for months, only when I had Anchovies the other day did I realize. Anchovies has a mega high percentage of Purine in them, which is a major cause of Gout in the body. So the Gout that was slowly receding, it immediately flared up, and got super bad again. My toe swelled up, and it hurts just like Gout symptoms. This also explains why I kept getting flare ups everytime I took multivitamins due to the B vitamins in them (Niacin). Considering I am healthy, never had Gout or other akin sicknesses, NMN is the almost 100% proven cause of the onset of this sickness. Tried to get a health boost, listened to Snake Oil salesmen on RU-vid, and boom, now I could have ongoing issues with this sickness. Gee thanks... Lesson learned! I recommend not taking NMN and focusing on other longevity protocols. I feel it has become super clear now that David Sinclair is just another Snake Oil salesman. His authentic demeanor is just a good mask in my opinion.

And taking it can cause Vitamin B overdose, causing horrible symptoms, some of which I saw. At this point, it is very clear that David Sinclair is just another of the many charlatans out there.
Dr Stanfield, Excellent. 1. I may switch to Naicin, but one big was not covered, that is mouth disolved NMN that never goes into the gut. That is way I use it. Any info on that, have to think it would be different? and 2. on Naicin there are many things believed to help with artery deposits, such as Nattokinease, K2-MK7, L-Argintine, etc. The B3 Flushing aspect might be a positive for increased blood flow for people with artery issues it would seem. Your thoughts?

Yes your arguments are compelling, the other thing I worry about is boosting NAD through supplementation may lead to the body to adapt in producing less NAD naturally. If you don't remember to regularly supplement they could be perverse outcomes.
It won't, but it could lead to a large amount of reduced NADH, which could be detrimental. You need to restore the NAD metabolic pathways, not just ram more NAD into your system.

@@Nelis1324 There is a way to restore NAD+ production to normal levels, restore mitochondrial complex 1 to 4 and atp levels at the same time as restoring NAD+ to NADh balance to normal, youthful levels. It's cheap and easy, removes fat from the liver, and restores some level of Nucleotide synthesis in the process. I am somewhat reticent to say, because I'm working on patents, though. It's an alternative to NMN. it's cheap, easy, and natural. Lots of potential to make money from a novel mixture with it in it. I'll give you a hint. It takes advantage of how the body switches from Oxphos to Glycolysis, and back again. And it's slightly downstream from how exercise causes benefits, lactate production; but without the cancerous potential of lactate.
Flushing aside, niacin is also hard on the liver(look up studies) and since there currently is no guarantee of any longevity benefits, it's better to be safe now, than sorry later on! 😉
@@nozhki-busha Yes, initially I thought it was just that simple, but no, the quick release version is also hard on the liver, but in a dose dependent manner of course. And some people's liver can tolerate more than others. But I seen examples of the problem starting with people using above 70mg doses of the quick release version. This is also why people on statins(or other drugs that affects the liver) should be extra carefull taking niacin/nicotinic acid! 😉

Interesting video. I suppose you wouldn't expect an increase in NAD muscle levels for an old person that exercises regularly. His NAD levels as you pointed out are high already. So why look into NAD levels after supplementation unless you decide to test the old population with no exercise activity. There are organs that I am interested in that would be important to see. The eye, especially the retina may benefit from increased NAD levels. There are many age-related diseases that may benefit from supplementation. The assessment of retinal function and structure are now at impressive levels of detail. I suggest keeping the curiosity and interest in an area that has already shown some anti-aging effects and concentrating on tissues that have an unmet need for this recovery.
I agree. And most important: diet. Age related problems are caused by sugar and seed oils! Taking supplements make no sense if we do not take away the things that cause the illnesses. So stop sugar and seed oils and then exercise and take supplements.
I tried NMN and I even still have like 20 grams of it but it gives me nothing compared to what I get from taking B3 Niacin (Nicotinic Acid), this gives me way more energy and good feel!
@@Fearzero Anecdotal yes, placebo is pure hypothesis from your part. I know it's not placebo because you will feel your entire body being filled with warmth and energy, due to your bloodvessels expanding (flush effect) so calling that placebo is nothing but ignorant hypothesis from your part. And just because something is anecdotal does not mean the observations from anecdotes cannot be used scientifically.

the study he mentioned about NR was of middle-aged, obese, insulin-resistant men. As he mentioned in the beginning of the video, lack of exercise might decrease NAD. And I believe obese middle-aged men constantly have their NAD levels dropping. So the fact that by the end of the study their NAD levels remained the same might actually be a good thing, the NR coud've offset the NAD levels they are losing by virtue of being obese. For animal studies, NR and NMN have both shown to increase NAD in muscles. In a study on mice, NR supplementation increased NAD+ levels in skeletal muscle. Another study on mice found that NR supplementation increased NAD+ levels in the quadriceps muscle. In a study on mice, NMN supplementation increased NAD+ levels in the blood and skeletal muscle. Another study on rats found that oral administration of NMN increased NAD+ levels in the blood and gastrocnemius muscle. Proper human studies are needed, ideally not obese humans.
I noticed that a couple of the nicotinamide trials you mentioned were topically-applied forms that improved the condition of the skin. Is it fair to conclude these benefits can be derived from ingesting it as a supplement? Thanks for your work....
If NMN can actually promote longevity, why is it not tested in the Intervention Testing Program of the NIA, which is the gold standard of aging research. Are the NMN companies afraid that NMN will fail in the study just like NR?
Best summary yet. Someone should corner Charles Brenner on one of these vids and demand he answer the basic Q--when is he going to determine whether his NR ever gets beyond the bloodstream and into an actual cell. Brenner dances around this Q--although, how difficult would it be to determine???
I am yet to try NMN though i am very close to taking the plunge, however i have read tons of anecdotal benefits from forums etc. Sure that's not scientific but for argument's sake let's just assume that these benefits are real. Are you saying that these same benefits would also come from Nicotinic Acid? OR, could it be another action of NMN etc that is causing these benefits rather than them increasing NAD+ in muscles which they purported don't do.
I have been taking NMN from DoNotAge with TMG started with 500 mg then raised to 1000mg. 500 had no affects at all. 1000mg actually made me feel worse. I stopped and gone back to niacin. I have been execrecsing regularly, I am on a super clean healthy diet, maybe NMN only works on certain combination no idea. Did nothing for me.
It works. I have been taking it for a year and my kidney function has improved (I only have one kidney) my energy levels are much higher, and overall I just feel so much better. NMN and Magnesium have been the best supplement I have ever used, and by that I mean a noticeable difference. I had horrible headaches for a very long time and Magnesium had almost completely solved that problem. After the addition on NMN I feel like a completely different person compared to how I felt 2 years ago.

I've got a question about taking nicotinamide (aka niacinamide). Although it's a bit different from niacin, which you're taking 50 mg of a day, most nicotinamide products on Amazon have a dose of 500mg. Do you think it's far too high a dose to take every day? Also, there are large-scale clinical trials going on in the UK and Australia that give glaucoma patients 1.5g to 3g per day of nicotinamide. If these high does nicotinamide trials were to turn out to be effective against glaucoma, would it be okay to take such high doses of nicotinamide?
@Jaded K I think Chris Masterjohn said that 31 mg of NA is about what's needed. And that if you take too much the body has to get rid of it. That spends Methylgroups! Lack of Metylgroups can become serious. You can avoid that by taking TMG, which supply them, if you take much of NA. I suspect that the research you mentioned didn't care about Methylgroups. If not, maybe they don't really understand what they are doing. My multivitamin has 73 mg of Niacinequvivalent in the form of NA. That should be enough, I think. I experimented with 300 mg of NR and 500 mg of TMG. No discernible effect. So, I'm back to just my multivitamin.

I’ve used their NMN complex before going out drinking during the holidays and anecdotally, I can say for certain that it cures any hangover you might get the next day. No matter how much I drink or if I slacked off on water hydration, I always felt shockingly robust and energetic the following day. That may not be the way for everyone, but at least I know it’s doing something extremely useful.
As they age, each mitochondrion accumulates nadh to the point where it seems to bog down energy production. That is a part of mitochondria aging and nmn or any b3 seems to rescure this process. But it's not the only thing that ages mitochondria, and mitochondrial aging is only one aspect of aging. Fasting will help a greta deal more with aging but even with extreme fasting I doubt anyone is going to find they live to 130 because of their lifestyle and diet, it's just not realistic. But it will keep you from dying off as early from chronic disease. Some of the many benefits of doing occasional extended fasting: Blood clotting is reduced and blood clots and arterial plaque are reabsorbed into the body. Blood pressure is quickly and dramatically lowered. Fribrosis/scarring is reversed over time and telomeres are lengthened, which also helps with lung fibrosis. Reflexes and short term memory are increased. Fasting increases nitric oxide. Fasting stimulates phagocytosis, the ingestion of bacteria, plaques and viruses by the immune system. Vitamin D plasma levels are increased, and vitamin D in turn increases autophagy! Fasts from 36-96 h actually INCREASE metabolic rate due to norepinephrine release! Weight loss from fasting only loses10% lean tissue and 90% fat compared to the typical 25% lean tissue and 75% fat lost when calorically restricting for long periods. The hunger hormone ghrelin lowers with extended fasting and rises from dieting. Blood sugar and insulin are lowered, allowing white blood cells to move more freely throughout the body and do their job. Ideal blood sugar is around 80. Some viruses activate glycolosis (the release of sugar in the body) and clinically it has been shown that decreasing glucose metabolism in the body weakens the influenza virus. Thymus is regenerated, which suppresses aging and renews the immune system. When you move out of MTOR your body shuts down the building blocks of the cell which are used to produce organelles and proteins. This means the mechanisms needed by viruses to replicate are by and large unavailable when you are in a deeply fasted state. What breaks a fast? Anything with protein or carbohydrates in it will break a fast, though if the amount is tiny you will go back into ketosis very quickly. Most teas and herbs are OK. Most supplements and meds will either break ketosis directly or contain a filler that will. Many medications are dangerous to take while fasting so you may have to talk to your dr. about discontinuing them during a fast. When you fast, this stimulates apoptosis in senescent or genetically damaged cells. This kills these cells off completely. Senescent cells are responsible for the effects of aging and are the root cause of the development of cancer. If it were possible to destroy them all it would completely stop aging and cancer. That is not possible but fasting can help limit these effects by killing off many of the affected cells and limiting the future effects of aging. Fasts of several days will not affect short term female fertility and may increase long term fertility, especially in women with PCOS. Does fasting lower testosterone? No, it raises it when the fast is broken by increasing lutenizing hormone and helps build muscle by increasing insulin sensitivity! The hormone Leptin is an immunomodulator that keeps the body from attacking itself and obesity causes leptin resistance. Fasting very quickly reduces leptin resistance and leptin levels and one day of fasting can cut your leptin levels in half and gets your immune system working properly again! Does the body preferentially prefer glucose as a fuel? No, your body always runs mainly on fat except for brief periods of very intense exercise. Your brain also prefers to burn ketones at a rate of around 2.5 to 1 when they are available in equal quantity to glucose. Fasting stimulates the AMPK complex and activates autophagy. Autophagy (literally self eating) will cause cells to recycle foreign matter such as viruses and kill cancerous and senescent cells. AMPK does many helpful things in the body including activating the body's antioxidant defenses. Deep ketosis virtually eliminates chronic inflammation in the body. This can offset the life threatening symptoms of viral pneumonia which effectively kills you through inflammation. This also creates BHB ketones in your body, which also help your immune system and anti-oxidative system, especially in the brain. Ketones also provide an additional energy source during infection, which is critical when trying to fight off a bug. In fact you can have as much as three times the total energy available in your blood when you are in deep ketosis, or even more. It increases mitochondrial function and repairs mitichondrial DNA, leading to improved ATP production and oxygen efficiency and thereby making cells better able to fight off infection. Increased mitochondrial function also has the added benefit of increasing your metabolism and cancer prevention! Fasting also releases BDNF and NGF in the blood which stimulates new nerve and brain cell growth, helping a great deal with diseases like MS, peripheral neuropathy and Alzheimers. In fact, the biochemical regulator of BDNF production is beta-hydroxybutyrate, which is the same ketone the body produces to nourish the brain while fasting. Fasting also increases telomere length, negating some of the effects of aging at a cellular level. A fasting mimicking diet for 3-5 days in a row also provides many of the same benefits as water fasting. FMD usually has 200-800 calories, under 18 g of protein and under 18 g of carb. Exogenous ketones can aid with fasting, making it easier in healthy people and allowing some people with specific issues to fast in spite of them without worrying as much about hypoglycemia. Children, pregnant or nursing women should not fast for periods longer than 16 hours. People with pancreatic tumors or certain forms of hypoglycemia generally cannot fast at all. Type 1 diabetics can also fast but it is more complicated and should be approached with caution as it could lead to ketoacidosis. Those with Addison's disease may also be unable to fast without liberal use of exogenous ketones, depending on severity. There's a variety of B3 that hasn't been mentioned: inositol hexanicotinate It's a slow-release form of B3 which works better than flush B3. It's been used for years by doctors like Abram Hoffer. ~500 mg seems to be a sweet spot, but 250 mg also seems to work. N.B. Using only 100 mg (a B-complex tablet) seems too little to get all the benefits Hoffer regularly used higher amounts of 3 g or more, but the desirable dose probably depends on the problem you are trying to address: Your blood tests should help you titrate the dose to optimise the benefits. 🙂
Excellent Vid Brad. I too was hugely excited after reading David Sinclair's book and rushed off to buy a bag full of NMN which is essentially how I came to your youtube channel. Unfortunately, after taking it for a month I tried to find out more about the company that supplied me and discovered that they were connected to a dubious business person and no further info was available on their product. Ho Hum I thought. Your chart, the one you showed earlier with the various versions of the Vit B became my guide and led me to the lovely cheap Niacin. I do worry that this subject hasn't gone away yet and we shall be seeing a re-visit to this vid subject but for me I am happy not to be taking it.

Good man - appreciate this. The hype was concerning given the excellent and proven properties of niacinamide. There's some very interesting research rabbit holes to dive down when you consider optimising glucose availability, niacinamide and generally strong thyroid performance and anti-aging. All very interlinked in the Krebs and in cell/tissue repair. Baffling to me why more research $ aren't spent on this kind of "energetics" research.
It's in the interest of the medical/pharma industry to have you unhealthy and in need of medicine for acute maladies, and unfortunately they have enough sway over relevant federal bodies to discourage any robust research into how we can stay healthy and avoid this giant grift.
One thing which I would add is that so many doing "reviews" are actually paid by the companies, and/or cashing in by marketing on RU-vid, etc. When there's money involved, I'm always super sceptical. However, there is a lot of supporting science, and my curiosity got the better of me, so I'm trying NMN to see if there are any results. I'll give it a few months. I had tried high dose B3 before, but nothing whatsoever. I often have tiredness, which I've suffered from all my life. As a teenager I'd get very sick just doing some physical activity, actually vomiting. Tired so much, and have seen little improvements, but nothing as dramatic as the claims for NMN. So, we'll see.

Please do one on NADH. None of the NAD researchers or commentators say anything about it. Yet it has a following. One small study found it effective for me/cfs..which I have.
Is there anything I can take with alcohol before during after that will help prevent my body converting alcohol to alacitate or whatever that poison is. Like I feel like I need to keep drinking because of stress in my life and just wanting to disconnect and forget or zone out but still wanna be healthy and not get cancer so yeah looking at NMN at the minute but yeah heard of DNA repair products and others yeah call me silly but that's me
There's been a few human studies which have not only shown increased NAD+ blood levels but also better muscle endurance in humans. i don't think that came out in this video. Also, if multiple studies show that blood NAD+ levels are increased, then repeating the conclusion over and over as you did, that NMN is probably absorbed and converted into other forms of B3 anyways is not supported by the data.
I think a critical piece of info missing in this analysis and many studies is that NMN was not studied alongside low dose resveratrol (less than 250mg in my non-expert opinion). The two are synergistic and NAD+ levels may not go up as much without taking both. (Note that high dose resveratrol can be detrimental but studies suggest low dose is very beneficial. Also, it seems some recommend one day on, one day off for resveratrol.)
So if it hasn't been studied, how do you know they are synergistic? I've heard it too but I would like to know why people are saying it. Is it just in theory?
I'm not knowledgeable in biology. If NMN supplements, as you state at 4 :00 "do boost blood NAD", what would be the explanation for why it doesn't affect the muscles? Does not the blood enter the muscles? What biological process would intervene between the circulating blood having increased NAD but not the muscles? Thanks.
I keep trying to decide if i need this and what to use? It is confusing and when looking at online info for B-3 the “may help” is ever present when talking about effects! So is it better to use 50mg of b3 or NAC?
